Objective: By testing the change of concentrations of hepcidin(HEPC),von Willebrand factor(vWF),angiotensin ?(Ang ?)and lectin-like oxidized low density lipoprotein receptor-1(LOX-1)in blood before and after donation,effects on donation of blood on HEPC,vWF,Ang ?,and LOX-1 were systematically investigated.And their clinical values on potential iron storage status,vascular endothelial damage status,oxidative stress state and lipid peroxidation state were therefore explored.Object and Methods:70 blood samples were collected from the voluntary blood donations from June 2017 to December 2017 in Tianjin.20 volunteers without blood donation were selected as control group.Strict medical examinations for all participants were conducted prior to blood donation in accordance with the health examination standard for blood donors issued on 30 December 2011(GB18467-2011).Methods: The volunteers were selected to donate more than 2 times as blood donation group,low frequency blood donation group: blood donations of 2 times,intermediate frequency blood donation group: blood donation of 3 to 9 times,high frequency blood donation group: 10 or more blood donations,20 volunteers without blood donation as control group(four groups of blood donation).A total of 29 volunteers were selected as?5 blood donation group.A total of 41 volunteers were selected as <5 blood donation group(two groups of blood donation).1.Enzyme linked immunosorbent assay(ELISA)was used for HEPC,vWF concentrations.Baseline data,HEPC and vWF were compared between the four groups and two groups of blood donation.In comparison,single factor,multivariate Logistic regression analysis and ROC curve analysis were performed.2.The ELISA was also used for detection of the concentration of Ang? and LOX-1.HEPC and vWF were compared between the blood donation group and the control group.Logistic regression was used for analyzing the influence factors of blood donation on the state of oxidative stress and lipid peroxidation.ROC curve was used for evaluation the diagnostic power of Ang?and LOX-1 for the state of oxidative stress and lipid peroxidation.Kaplan-Meier probabilistic analysis was used for analyses of low,medium and high frequencies three blood donation groups with the increase of frequency of blood donation,the probability of potential oxidative stress state and lipid peroxidation state.Results:1.Compared with the control group,the HEPC concentrations of the high frequency blood donation group decreased significantly.The vWF concentrations of high frequency and middle frequency blood donation group were significantly reduced with statistically significant(P<0.05).In the range of high frequency blood donation group and the control group,Logistic regression showed that the declines of both HEPC and FER,vWF and FER were protective factors after high frequency blood donation.ROC curve analysis showed that the decrease of HEPC concentrations had a certain diagnostic effect on the improvement of potential higher iron storage condition,the decrease of vWF concentrations had certain diagnostic effect on the improvement of potential vascular endothelial cell injury.The concentrations of HEPC,vWF,FER in the ?5 blood donation group were significantly lower than that of <5 blood donation group.In the range of ?5 blood donation group and <5 blood donation group,Logistic regression and ROC curve showed that,the decline of HEPC and FER,vWF and FER were protective factors after five or more blood donation,and the collaborative application of HEPC and FER had certain diagnostic value on the improvement of the potential higher iron storage state.The collaborative application of vWF and FER had certain diagnostic value on the improvement of the body potential vascular endothelial damage status.2.The Ang ? and LOX-1 concentrations of blood donation group were significantly lower than the control group,with statistically significant(P<0.05).The Logistic regression analysis showed that the decline of Ang? concentrations and LOX-1 concentrations were the protection factors after many times blood donation.ROC curve analysis showed that Ang ? concentrations decrease had certain diagnostic efficacy on the improvement of the body potential oxidative stress state,LOX-1 concentrations decreased had certain diagnostic efficacy to improve the potential of lipid peroxidation.The Kaplan-Meier probabilistic analysis of low,medium and high frequencies blood donation groups showed that with the increase of blood donation frequency,the probability of potential oxidative stress and lipid peroxidation decreased.Conclusions:1.The declines of HEPC,vWF,Ang ? and LOX-1 concentrations are the protection factors after many times blood donation.They can promote the improvement of the potentially poor iron storage state,vascular endothelial damage status,oxidative stress state and lipid peroxidation of the body.
