Objective: To investigate the effect of different concentrations of uric acid on Human renal glomerulus endothelial cells(HRGEC)injury associated factors.Methods: HRGEC were cultured in vitro,stimulated by different concentrations of uric acid(0,4,8,16mg/dl)for 24 hours,the microscope was used to observe HRGEC,the cell counting kit was used to test the activity of cells,Real time PCR was used to test the expression of IL-1?TNF-? ?SOD2.Results:1.Compared with blank control group(the concentration of uric acid was0mg/dl),the expression of IL-1 m RNA of both physiological concentration of uric acid group(the concentration of uric acid was 4mg/dl)and high concentration of uric acid group(the concentration of uric acid was both 8mg/dl and 16mg/dl)was markly increased,the difference was statistically significant(P<0.05);Compared with physiological concentration of uric acid group(the concentration of uric acid was4mg/dl),the expression of IL-1 m RNA of high concentration of uric acid group(the concentration of uric acid was both 8mg/dl and 16mg/dl)was markly increased,the difference was statistically significant(P<0.05);Compared with the 8mg/dl of uric acid,the level of IL-1 m RNA was markly increased when the concentration of uric acid was 16mg/dl,the difference was statistically significant(P<0.05).2.Compared with blank control group(the concentration of uric acid was0mg/dl),there was no significant difference in the expression of TNF-? m RNA of physiological concentration of uric acid group(the concentration of uric acid was4mg/dl);the expression of TNF-? m RNA of high concentration of uric acid group(the concentration of uric acid was both 8mg/dl and 16mg/dl)was markly increased,the difference was statistically significant(P<0.05);Compared with physiological concentration of uric acid group(the concentration of uric acid was4mg/dl),the expression of TNF-? m RNA of high concentration of uric acidgroup(the concentration of uric acid was both 8mg/dl and 16mg/dl)was markly increased,the difference was statistically significant(P<0.05).3.Compared with blank control group(the concentration of uric acid was0mg/dl),the expression of SOD2 m RNA of physiological concentration of uric acid group(the concentration of uric acid was 4mg/dl)was markly increased,the difference was statistically significant(P<0.05);the expression of SOD2 m RNA of high concentration of uric acid group(the concentration of uric acid was both 8mg/dl and16mg/dl)was markly reduced,the difference was statistically significant(P<0.05).Compared with physiological concentration of uric acid group(the concentration of uric acid was 4mg/dl),the expression of SOD2 m RNA of high concentration of uric acid group(the concentration of uric acid was both 8mg/dl and 16mg/dl)was markly reduced,the difference was statistically significant(P<0.05).Conclusions:1.High concentration of uric acid may activate inflammatory reaction to damage glomerular endothelial cells.2.Physiological concentration of uric acid has antioxidant activity,but high concentration of uric acid may activate oxidative stree mechanism to damage glomerular endothelial cells.
