Object: The aim of the study is to evaluate the endometrial receptivity in infertile women with endometriosis at clinical,molecular,cell,protein and gene levels during the window of implantation and to explore the mechanism.Methods: 1.Patients with endometriosis infertility(n=24)were selected as endometriosis group and fertile women without endometriosis(n=14)were selected as control group.The folliculars were monitored by ultrasonography from the ninth day of the menstrual cycle.During 6-10 days after ovulation(the LH test showed double line and the ovulation sign was detected by B ultrasonic examination),serum and eutopic endometrium tissues were collected.Serum levels of estrogen and progesterone were measured by radioimmunoassay(RIA);Endometrial thickness,endometrial types(A,B,C),subendometrial blood flow(0,I,II,III)was detected by abdominal and transvaginal ultrasound.2.Scanning electron microscope to observe pinopode,cilia and microvilli on endometrial surface;Immunohistochemical analysis,RT-PCR and western blot to detect the expression of endometrial receptivity markers(?V?3 integrin,MUC-1,HOXA10 and LIF).3.Lnc RNA expression profiles by human lnc RNA Array v4.0.QRT-PCR method was used to verify differentially expressed Lnc RNA;In situ hybridization location experiment was applied to locat the lnc RNA;Target mi RNA and m RNA were analyzed by bioinformatics and then detected by QRT-PCR.Result:1.There was no significant difference in the the levels of estrogen and progesterone,thickness of endometrium,endometrial type(A,B,C)and subintimal blood flow(0,I,II,III)during the window period between the endometriosis group and control group.2.The two groups of eutopic endometrium histopathologically showed normal intima tissue;The surface of the endometrium lacks pinopode,cilia and microvill in infertile women with endometriosis.There were statistically significant differences in pinopode coverage and in the proportion of cilia and microvilli between endometriosis group and control group(P?0.05).The expression of ?V?3 integrin,MUC-1,HOXA10,LIF were significantly decreased in endometriosis group compared with the control group by q RT-PCR and immunohistochemistry.The protein expression of LIF was significantly decreased in endometriosis group compared with the control group by western blot.3.A total of 945 lnc RNA were up-regulated and 830 lnc RNA down-regulated in the endometriosis group endometrium compared with the control group endometrium by lnc RNA microarray;The expression of lnc RNA NR-121610 was significantly decreased in the endometriosis group.In situ hybridization showed that it was mainly located in the cytoplasm.The expression of has-mi R-580-3p related to lnc RNA NR-121610 was significantly upregulated in infertile women with endometriosis by q RT-PCR.The expression of has-mi R-580-3p was negatively correlated with ?V?3 integrin m RNA expression.Conclusion: 1.Endometrial receptivity in endometriosis associated with infertility patients is decreased.2.The abnormal expression of lnc RNA NR-121610-has-mi R-580-3p-?V?3 integrin of infertile patients with endometriosis is closely related to the decrease of the endometrial receptivity.
