Objective:1.Establishing a mouse model of endometriosis;2.Detecting the protein expressions of HOXA10 and SUMO1 in eutopic endometrial tissue of mice by Immunohistochemistry after dividing mice into different groups randomly and taking interventions and the HOXA10-SUMO1 by Co-immunoprecipitation.Then the results were analyzed to explore whether the Bushen Huoxue Decoction can improve endometrial receptivity of Endometriosis by affecting SUMO1 modification of HOXA10.Methods:Part ?Looking up and reviewing the relevant studies.Part ?1.Choosing mice with a regular estrus cycle according to vaginal exfoliated cytology.2.Establishing endometriosis mouse model of BALB/c by intraperitoneal injection.3.Verifying the success rate of endometriosis mouse model on day 7 and 21.4.After day 21,dividing the mice into model group,Bushen Huoxue Decoction group and sham operation group.5.Mice in the Bushen Huoxue Decoction group were given Bushen Huoxue Decoction for 30 days,both model group and sham operation group of mice were given an equal volume of distilled water.6.After gavage,the uterus of mice in each group were taken,one part was put into tissue fixative for preservation,and the other part was preservated in-80?.7.Detecting the protein expressions of HOXA10 and SUMO1 in eutopic endometrial tissue of mice by Immunohistochemistry and the HOXA10-SUMO1 by Co-immunoprecipitation.8.Analyzing the results Statistically,gussing whether the Bushen Huoxue Decoction can improve endometrial receptivity of endometriosis by affecting the protein expressions of HOXA10 and SUMO1,and SUMO1 modification of HOXA10.Results:1.The success rate of establishing endometriosis mouse model by intraperitoneal injection was 100%;2.According to the immunohistochemical results,the expression of protein HOXA10 between Bushen Huoxue Decoction group and the sham operation groups was not significantly different((P>0.05),while it significantly increased in the model control group(P<0.01),the expression of protein SUMO1 in the Bushen Huoxue Decoction group and the model control group increased abnormally,especially in the model control group(P<0.01).The results of co-immunoprecipitation showed that the expression of protein HOXA10-SUMOl in the Bushen Huoxue Decoction group was significantly lower than that in the model control group.3.The expression of protein HOXA10 and SUMO1 in Bushen Huoxue Decoction group were positively correlated with moderate intensityConclusion:1.The expression of protein HOXA10 decreased and protein SUMO 1 increased significantly in endometriosis model mice.2.The Bushen Huoxue Decoction can increase the expression of protein HOXA10 and slightly decrease the expression of protein SUM01,It can also decrease the expression of protein HOXA10-SUMO1,Thus,Bushen Huoxue Decoction may improve the endometrial receptivity of endometriosis by inhibiting the SUMO1 modification of HOXA10.
