This paper studied on the identification and quality analysis of two kinds of Tujia medicinal materials used in western Hubei hospital,and research was divided into two parts.The first part was about the study of the Nandinae caulis,including the pharmacognosy identification,the determination of total alkaloids,the study of HPLC characteristic chromatogram and the determination of some active substances.The second part is about the study of Viticis negundinis caulis from the two origin plants(Vitex negundo and Vitex negundo var.cannabifolia(Sieb.et Zucc.)Hand-Mazz.),which includes pharmacognosy identification,HPLC characteristic chromatogram and the content determination of some active substances.In the middle stage of the study,according to the deadline of the related subject,the quality standard draft for two kinds of medicinal materials have been drawn up(excluding content determination due to the time limitation).The Nanadinae culis is the dried stem of Nandina domestica Thunb.which has the functions of clearing heat,dehumidifying and relieving cough,and was recorded in "Chinese Materia Medica" with the name of Nanadinae culis and many other pharmacological monographs.Field investigation showed that the medicine was used in county level Chinese medicine hospital of western Hubei for many years,but there was no quality standard for it.The pharmacognosy identification and related examination items were studied,including the macroscopic and microscopic characteristcs,TLC identification methods,and the investigation of water content and extracts,etc.And the draft of quality standard of Nanadinae culis was written.In the later period,the determination method of the total alkaloids was established,and the extraction conditions,the kinds and amounts of the acid dyes,and the pH value of buffer were optimized.At the same time,the HPLC fingerprint chromatogram and the content determination method of some active substances were researched,and the differences of the samples from different producing areas and different harvest periods were compared,and the relative regularity of the quality of medicinal materials was preliminarily analyzed.The results of pharmacognosy identification showed that the older stem had the characteristics of cork tissue connected to phloem and phloem fiber group arranged in the shape of "U" or annular.In the TLC identification results,there were five clear and consistent main fluorescent spots on the chromatogram,and the yellow-bright spots corresponded to the spots of berberine hydrochloride.The results of fingerprint chromatogram and content determination showed that the samples from Wufeng County and Xianfeng County had significantly higher contents,while the other samples had little difference.It was found that the sample rings in Wufeng County and Xianfeng County were mostly 5~7 cycles,while those in other producing areas were mostly 2~4 rings.In the samples of different harvest periods from three producing areas,the content of the components did not change with the month of collection,but increased slightly with the time of harvest.It is inferred from the above results that the factors of different producing areas and different harvest periods had a limited effect on the content of the components of this medicinal material,but the number of growth years had a great effect on it.It was preliminarily judged that the content of chemical components in this medicinal material was on the rise with the increase of growth time.Viticis negundinis culis is the dry stem of Vitex negundo L.or Vitex negundo L.var.cannabifolia.(Sieb.et Zucc.)Hand-Mazz which has the function of dispelling wind and relieving pain."Chinese Materia Medica" recorded two kinds of medicinal materials were Vitex negundo brunch and Vitex negundo stem,and its main treatment functions are basically in accordance."The standard of Chinese medicinal materials in Fujian"(2006 Edition)recorded Vitex negundo var,but the description of its characters is too simple,and the description of microscopic characteristics is only consistent with the tender stem sample which is different from the actual application of the coarse stem.In this study,a more systematic pharmacognostic identification method was established based on the actual medicinal materials,the limited value of the relevant inspection items and other routine items in the standard were worked out,and the quality standard draft of the medicinal material was formulated.In the later period,the HPLC fingerprint chromatogram of the medicinal material was studied,and the chromatograms of the samples from different base sources was compared.During the study,the chemical constituents were analyzed by LC-MS,and some active components were determined by the method of content determination.The results showed that because of the high affinity of the basic plants,the characters,microscopes and TLC of the two species were consistent,and their HPLC chromatogram were basically the same(The chemical composition of the two is basically the same).In combination with the clinical application,it is reasonable to use the two basic sources as the source of the Viticis negundinis caulis.The results showed that in the sample of Snake Mountain in Wuhan,with the development of harvest time,the content of components of the sample with base source was obviously increased,and the sample with base source of Vitex negundo also showed a slightly upward trend.From the above results,it can be seen that the content of chemical components in this medicinal material increased following the extension of growth time.As a supplement,the author have compared the infrared spectra of two species of Viticis negundinis caulis,which further proved the rationality of two sources of Viticis negundinis caulis.The methods of identification and quality analysis of two kinds of medicinal materials were established in the study,and the quality standards of two kinds of medicinal materials have been drawn up(is going to be recorded in "Quality Standard for Chinese Medicinal Materials in Hubei",2018 edition).It provided a basis for the identification and analysis of the safety and effectiveness of these two kinds of medicinal materials.The determination of the content of related active components in the samples of two kinds of medicinal materials from different producing areas and different harvest periods showed that with the delay of harvest period or the extension of the growth years of the medicinal materials,the content of the active substances increased significantly,and it was suggested that the content of the active substances in the samples increased significantly with the increase of the growth years of the medicinal materials.As a stem medicinal material derived from perennial woody plants,the total active substances should be gradually increased with the extension of growth time or growth year in a certain time range.
