Periodontal ligament as an unique anatomical structure can response to mechanical and inflammatory signals independently,and induce the continuous bone deposit and bone resorption in local alveolar bone.Orthodontic stress may not induce the loss of bone mass,but inflammatory factors can induce the continuous bone resorption.The Eph/phrin interaction serves as a bidirectional signaling.More details on the regulation of osteoblastic and osteoclastic differentiation by Eph and ephrin family members have been discovered recently.In this study we plan to investigate the effects of inflammatory factors on hepatocyte kinase receptors and ligands in human periodontal ligament cells.Methods:In this research,Human periodontal ligament fibroblasts are stimulated with inflammatory cytokines(10 ng/ml TNF-a,10 ng/ml IL-1β).The expression of Eph and ephrin at both mRNA and protein levels are examined.Results:Two inflammatory factors caused similar changes in the expression of Eph receptors and ephrin ligands in periodontal ligament cells.The expression of ephrinA2 increased in a time-dependent manner,while the expression Eph4 receptor gradually decreased after a transient high expression.Conclusion:Both TNF-a and IL-1β can cause changes in expression of Eph receptors and ephrin ligands in human periodontal ligament cells,and the expression changes induced by both inflammatory factors are consistent,but the effect of TNF-a is more pronounced.
