Objective In recent years,the incidence of colon cancer worldwide has been increasing and colon cancer is one of the most common gastrointestinal tumors.It still lacks effective treatment and has a high recurrence rate.In order to explore new biomarkers and therapeutic targets related to colon cancer,it is very important to study the molecular mechanism of colon cancer.The role of non coding RNA(non-coding RNA,ncRNA)in colon cancer has attracted much attention.This study focused on the expression profile of RNA micro RNA(miRNA)in colon cancer tissues.Methods Firstly,we collected 5 cases of colon cancer and 5 cases of adjacent normal colon tissues from the Endoscopic Center of Anhui Provincial Hospital.Total RNA was extracted with Trizol reagent,purified and quantified.Affymetrix microRNA 4.0 microarray and CapitalBio mRNA microarray were used to detect the genes,and differentially expressed miRNAs and mRNAs were screened out.Then the target genes of differentially expressed miRNAs were predicted by bioinformatics analysis,and the biological processes and signaling pathways enriched by target genes were analyzed by KEGG signaling pathway and GO analysis.Results In this study,73 deregulated miRNAs and eregulated 2459 mRNAs were screened in colon cancer by microarray assay(the criteria: Fold Change > 2 and corrected P < 0.05).Among them,22 miRNAs and 1464 mRNAs were up-reguleted in colon cancer,while 51 miRNAs and 995 mRNAs were down-regulated.The top deregulated miRNAs were summarized,including hsa-miR-183-5p,hsa-miR-1247-5p,hsa-miR-552-3p,hsa-miR-7-5p,hsa-miR-224-5p,hsa-miR-183-3p,hsa-miR-182-5p,hsa-miR-99a-5p,hsa-miR-133a-3p,hsa-miR-139-5p,hsa-miR-1,hsa-miR-143-5p,hsa-miR-149-5p,hsa-miR-30a-3p,hsa-mir-139 and others.In addition,based on deregulated mRNA,the target gene prediction analysis showed that miR-622 could regulate the expression of TGF-?,TP53,KARS,WNT,SMAD4,and miR-125b-5p could regulate the expression of STAT3,TP53,while APC was the common target gene of miR-125b-5p,miR-30a-5p,miR-149-5p,miR-18a-5p,miR-182-5p and miR-422 a.The protein modification,Wnt-signaling pathway,post-translational protein modification,cancer-related pathway,RAS signaling pathway and TP53 transcriptional regulation were enriched in colon cancer by GO analysis and KEGG pathway analysis.Conclusion The miRNAs play an important role in colorectal cancer.We screened 73 deregulated miRNAs and predicted target genes,enriched biological processes and signaling pathways.This will help us to deepen understand the gene regulation network and pathogenesis of colon cancer,and to provide a new perspective for searching the effective biomarkers of early diagnosis and therapeutic strategy.
