Objective In recent years,the incidence of colon cancer worldwide has been increasing and colon cancer is one of the most common gastrointestinal tumors.It still lacks effective treatment and has a high recurrence rate.In order to explore new biomarkers and therapeutic targets related to colon cancer,it is very important to study the molecular mechanism of colon cancer.The role of non coding RNA(non-coding RNA(ncRNA)in colon cancer has attracted much attention.This study focused on the expression profile of RNA micro RNA(miRNA)in colon cancer tissues.Methods Firstly,we collected 5 cases of colon cancer and 5 cases of normal colon tissues and 5 cases of colon adenoma from the Endoscopic Center of Anhui Provincial Hospital.Total RNA was extracted with Trizol reagent,purified and quantified.Affymetrix microRNA 4.0 microarray was used to detect the genes,and differentially expressed microRNAs were screened out.Then the target genes of differentially expressed microRNAs were predicted by bioinformatics analysis,and the biological processes and signaling pathways enriched by target genes of KEGG signaling pathway were analyzed by GO analysis.Results In this study,83 differentially expressed microRNAs in colon cancer were screened by Affymetrix microarray assay.Fold Change > 2 and corrected P < 0.05 were used as the test criteria.Among them,37 were over expressed in colon cancer,46 were overexpressed in colon cancer.110 differentially expressed microRNAs in colon adenoma were screened by Affymetrix microarray assay.Among them,53 were over expressed in colon adenoma,57 were overexpressed in colon adenoma.We summarize some of the most significant up-and down-regulated and down-regulated microRNANAs,including hsa-microRNA-183-5p,hsa-microRNA-183-5p,hsa-microRNA-1247-5p,hsamicroRNA-1247-5p,hsa-microRNA-552-3p,hsa-microRNA-552-3p,hsa-microRNA-microRNA-7-5p,hsa-microRNA-7-5p,hsa-microRNA-224-5p,hsa-microRNAmicroRNA-224-5p,hsa-microRNA-microRNA-183-183-3p,hsa-microRNA-182-182-5p,hsa-microRNA-182-182-5p,hsa-99a-Mi-143-5p,hsa-miR-149-5p,hsa-miR-30a-3p,hsa-mir-139 and other colon cancer related genes.In addition,target gene prediction analysis showed that differentially expressed target genes regulated by microRNAs,such as microRNAs,could regulate the expression of TGF-beta,TP53,KARS,WNT,SMAD4,STAT3,TP53,and APC,while microRNAs were also microRNAs-125b-5p,microRNAs-30a-5p,microRNAs-149-5p,microRNAs-18a-5p,microRNAs-182-5p,and microRNAs-42.The target gene of GO analysis and KEGG signaling pathway analysis showed that protein modification,Wnt signaling pathway,cancer-related pathway,post-translational protein modification,RAS signaling pathway,TP53 transcriptional regulation were involved in colon cancer.Conclusion MiRNA colon cancer plays an important role.Compared with normal colon tissue miRNAs,we screened 83 differentially expressed miRNAs in colon cancer and 110 differentially expressed miRNAs in colon adenomas.It also describes the target genes,enriched biological processes and signaling pathways regulated by them.This will help to deepen our understanding of the colon cancer gene regulatory network,and explore the pathogenesis of colon cancer,explore early diagnosis and effective biomarkers,and expand treatment strategies to provide new horizons..
