Objective: To study the culture of urine derived stem cells by collecting the middle morning urine of healthy adult male,and to make a biological identification of the urine derived stem cells.Methods: The urine of healthy adults was cleaned,the urine derived stem cells were obtained by centrifugation and separation,and the cells were cultured with attachment culture method.The growth morphology of the urine derived stem cells was observed under the inverted microscope,and the growth curves of the cells were measured by MTT,and the growth and change of the cells were observed.Cell phenotype was identified by flow cytometry.The cells were isolated,cultured and passaged.The osteogenic and adipogenic differentiation of cells were performed to observe the biological characteristics of human urinary stem cells.Results: In the urine,cells with homogeneous morphology,rapid growth and large proliferation were obtained.The cells grew on the wall.The initial cell morphology was short shuttle or round growth.With the extension of cell culture time,the cells gradually became a long spindle shape.The cells are scattered in the culture bottle,and the arrangement is relatively close.The human urine derived stem cells were cultured and passed on,and the growth curve of the cells was "S" line growth.Human urine derived stem cells have biological characteristics that differentiate into osteoblasts and adipocytes.After adipogenic induction in cells by oil red O staining under lipid droplets were red,indicating that cultured human urine derived stem cells to adipogenic differentiation ability in vitro;cells with osteogenic inducer,the alizarin red staining under the calcified nodules were red,indicating the in vitro culture of human urine derived stem cells have the ability to differentiate into osteoblasts.Conclusion: There is indeed a large number of proliferating cells in the urine of healthy adults.This cell is easy to separate and culture in vitro,and can differentiate into adipocytes and osteoblasts,and has strong proliferation and differentiation ability.The development of urine derived stem cells will open a new research direction for the regeneration and healing of organs and tissues of urinary tract defects.With the further research of human urinary stem cells,it will make an outstanding contribution to the development of regenerative medicine.The construction of urethral stents combined with urethral stem cells will also open a new era for medical cause.
