Objective:To investigate the Specific cytotoxic T lymphocyte cells(CTL)immune response and its clinical correlation and prognosis on tumor-associated antigen(TAA),New York esophageal squamous cell carcinoma antigen 1(NY-ESO-1),synovial sarcoma X break point gene 2(SSX-2),sal-like gene 4(SALL-4),melanoma-associated antigen A1(MAGEA1)and melanoma-associated antigen A3(MAGE-A3),in esophageal squamous cell carcinoma patients by enzyme-linked immunoblot assay(ELISPOT).To explore its application value in immunotherapy of esophageal carcinoma.Methods:A total of 51patients diagnosed with esophageal squamous cell carcinoma from April 2015 to October2016 in the Cancer Hospital affiliated to Xinjiang Medical University were collected.NY-ESO-1,SSX-2,SALL-4,MAGE-A1,MAGE-A3 specific CTL epitope peptides were used to stimulate peripheral blood mononuclear cells(PBMC)of esophageal squamous cell carcinoma patients.The frequency and intensity of CTL cells secreting interferon-?(IFN-?)were detected by ELISPOT,and its relationship between CTL cells with clinical features and prognosis of patients with esophageal squamous cell carcinoma was analyzed.Results:(1)The frequency of NY-ESO-1,SSX-2,SALL-4,MAGE-A1 and MAGE-A3 specific CTL reactions in peripheral blood of patients with esophageal squamous cell carcinoma was 19.6%(10/51)?9.8%(5/51)?29.4%(15/51)?27.5%(14/51)and 15.7%(8/51),and the response intensity was 36.33SFC/10~6 PBMCs?36.00SFC/10~6 PBMCs?78.00SFC/10~6 PBMCs?55.71SFC/10~6 PBMCs and 45.83SFC/10~6PBMCs;(2)The frequency of MAGE-A1 specific CTL reaction in N0-1 group was significantly higher than that in N2-3 group,and the difference was statistically significant(P=0.037);(3)19 cases of esophageal squamous cell carcinoma patients'overall 1-year survival rate after radiotherapy was 42.1%.The median OS was 9 months.The 1-year survival rate of MAGE-A1 and MAGE-A3-specific CTL positive patients was lower than that of negative patients.The median OS of positive patients was higher than negative patients,but there was no significant difference in total OS(P>0.05).Conclusion:The patients with esophageal squamous cell carcinoma can produce specific T cell immunoreactivity on tumor specific antigens NY-ESO-1,SSX-2,SALL-4,MAGE-A1 and MAGE-A3 polypeptides.To provide clues for immunotherapy of esophageal carcinoma.
