Objective: Exploring the expression and its effect in hypopharyngeal cancer to provide new targets for mechanism research and therapy for hypopharyngeal cancer.Methods:(1)Real-time PCR was used to detect the expression of MCM7 in human hypopharyngeal cancer cell line FaDu.(2)siRNA was used to knock down MCM7.siRNA-MCM7 group: using siRNA-MCM7 to treat FaDu cells.siRNA-NC group: using negative control siRNA to treat FaDu cells.We used Real-time PCR and Western blot to verify the expressed level of MCM7 in the two groups.(3)MTT assay and wound healing assay was carried out to detect the change of cell proliferation and cell migration ability.(4)In order to explore the effect of MCM7 in FaDu,we used TCGA database and bioinformatics methods to establish the co-expressed genes network of MCM7 in head and neck squamous cell carcinoma and using gene modular analysis to screen core gene modules.Results:(1)MCM7 was expressed in FaDu cell line.(2)The expression of MCM7 was significantly inhibited in FaDu by using siRNA.(3)The results of MTT showed that the cell proliferation of FaDu was significantly inhibited after knocking down MCM7(P<0.05).The results of wound healing showed that the cell migration of FaDu was significantly inhibited after knocking down MCM7(P<0.05).(4)The co-expressed genes network of MCM7 revealed that MCM7 may regulate DNA replication,DNA damage repair,cell cycle,cell proliferation in head and neck squamous cell carcinoma(P<0.05).The result of gene module analysis showed that MCM7 may play the role in proliferation and migration of FaDu through affecting the gene module including MCM6,RFC2,RFC5,CDC6 and so on.Conclusions:(1)MCM7 was expressed in FaDu.(2)The silence of MCM7 inhibited the cell proliferation and migration of FaDu.(3)MCM7 may play important roles on regulating the function including DNA replication,cell cycle,cell proliferation of FaDu through affecting the co-expressed gene modules.
