Objective: To investigate the effects of regulation of dynamin-related protein 1expression on myocardial protection of sevoflurane post-conditioning in diabetic rats.Methods: The experimental animals were randomly divided into 8 groups,6 in eachgroup.The first three groups were Wistar rats,including sham operation group,ischemia reperfusion injury group(I/R)and sevoflurane post-conditioning group(SPostC).The last five groups were selected GK diabetic rats,which were diabetic group(DM),diabetic ischemia-reperfusion injury group(DM+I/R),diabetic sevofluranepost-conditioning group(DM+SPostC),DMSO group,Mdivi group.A model of myocardial ischemia-reperfusion injury was established and inhaled 2.4%sevoflurane for 15 minutes before reperfusion.Mdivi-1 was intravenously injected into rats 15 minutes before ischemia to interfere with Drp1 expression.The plasma LDH level of each group was measured.The myocardial infarct size was measured and the myocardial ultrastructure was observed.The expression level of Drp1 in each group was observed by Western Blot.Results: Compared with the SPostC group,the myocardial infarct size of the DM+SPostC group was not obvious,the myocardial fiber arrangement was disordered,myocardial mitochondrial division was increased,and myocardial Drp1 expression was not observed.Compared with DM+I/R group,myocardial infarct size decreased,LDH level decreased,myocardial mitochondrialdivision decreased,myocardial fibrosis decreased,and myocardial Drp1 expression decreased.The transfer of Drp1 to mitochondria increased significantly after myocardial ischemia-reperfusion injury,and the number of mitochondria increased,indicating a significant increase in mitochondrial division.Diabetic rats were treated with Mdivi-1 after I/R,and Drp1 was transported to mitochondria and mitochondrial division was reduced.In addition,the myocardial infarct size was reduced after the inhibition of Drp1,and lactate dehydrogenase level were decreased.Conclusion: Mitochondrialdivision inhibitors reduce the expression of Drp1,which hinders mitochondrial division,reduces myocardial ischemia-reperfusion injury in diabetic rats,and restores myocardial protection after sevoflurane postconditioning.
