Objectives : To investigate the expression of CD138 in serum and tissues of patients with cervical cancer and its clinical significance. Methods:Serum and pathological specimens of 41 patients with cervical cancer who underwent surgical resection or biopsy before first radiotherapy and chemotherapy in Zhongshan Hospital affiliated to Dalian University from December 2016 to January2019 were collected.Serum and pathological specimens of 30 patients with benign uterine lesions who underwent hysterectomy at the same time were collected.Serum of30 healthy people who underwent physical examination at the same time were collected.Among 41 patients with cervical cancer,5 were under 40 years old,36 were over 40 years old,37 were squamous cell carcinomas and 4 were adenocarcinomas.According to the principle of treatment of cervical cancer,34 cases were classified as clinical stage≤II A,7 cases as clinical stage≥II B,25 cases as primary lesion≤4 cm,16 cases as primary lesion>4 cm;26 cases had no lymphatic metastasis and8 cases had lymphatic metastasis. The expression of CD138 and SCC-Ag in serum of cervical cancer patients, benign uterine lesions and healthy people was detected by enzyme-linked immunosorbent assay(ELISA).To analyze the relationship between CD138 and SCC-Ag and clinical stage and lymph node metastasis of cervical cancer patients, and the correlation between CD138 and SCC-Ag expression in serum of cervical cancer patients. The expression of CD138 and E-cadherin in benign lesions of cervical and cervical cancer tissues was observed by immunohistochemistry. The age, clinical stage, tissue type, tumor size and lymph of patients with CD138 and E-cadherin and cervical cancer were analyzed. The relationship between metastasis and the correlation between CD138 and E-cadherin expression in cervical cancer tissues.Results:1.The expression of CD138 and SCC-Ag in serum of patients with cervical cancer, patients with benign uterine lesions, and healthy people. To analyze the relationship between CD138 and SCC-Ag and clinical stage and lymph node metastasis of cervical cancer patients. The expression of CD138 and SCC-Ag in serum of patients with cervical cancerwas higher than that of benign uterine lesions and healthy people. The difference was statistically significant(P<0.05).The positive expression level of SCC-Ag in cervical lymph node metastasis group was higher than that in non-lymph node metastasis group, the difference was statistically significant(P<0.05).The expression levels of CD138 and SCC-Ag in serum of cervical cancer patients increased with the later clinical stage, but there was no significant difference(P>0.05).The expression level of CD138 in serum of cervical cancer increased with lymph node metastasis, but there was no significant difference(P>0.05).2.The correlation between CD138 and SCC-Ag expression in serum of patients with cervical cancer. In the serum of 41 patients with cervical cancer, the expression levels of CD138 and SCC-Ag increased, and there was a positive correlation between them(R=0.491).3.Expression of CD138 and E-cadherin in cervical tissues and its relationship with age, clinical stage, tissue typing, tumor size and lymph node metastasis of cervical cancer patients. The positive expression rate of CD138 in benign cervical lesions was 86.67%,and the positive expression rate in cervical cancer tissues was 24.39%.The positive expression rate of E-cadherin in benign cervical lesions was 90.00%,and the positive expression rate in cervical cancer tissues was 29.27%,the results showed that the expression of CD138 and E-cadherin in cervical cancer was significantly lower than that in normal cervical tissues, and the difference was statistically significant(P<0.05).The expressions of CD138 and E-cadherin in cervical tissues of patients with cervical cancer were not related to age, tissue type, tumor size, clinical stage, and lymph node metastasis, and the difference was not statistically significant(P>0.05).4.The correlation between expression of CD138 and E-cadherin in cervical cancer tissues.Of the 41 cervical cancer tissues,7 were positive for CD138 and E-cadherin,and 26 were negative for expression.E-cadherin was positively correlated with CD138 expression.(R=0.508).Conclusion:1.The serum levels of CD138 and SCC-Ag in cervical cancer patients were higher than those in benign uterine lesions and healthy control group. The expression of SCC-Ag in lymph node metastasis group was higher than that in non-lymph node metastasis group.2.There was a positive correlation between the expression of CD138 and SCC-Ag in serum of patients with cervical cancer.3.The expressions of CD138 and E-cadherin in cervical cancer tissues were lower than those in benign uterine lesions.4.CD138 was positively correlated with E-cadherin expression in cervical cancer tissues.5.CD138 was highly expressed in the serum of patients with cervical cancer and low expressed in the tissues of patients with cervical cancer, which indicates that abnormal expression of CD138 is related to the pathogenesis of cervical cancer.CD138 is expected to become one of the tumor markers in the adjuvant diagnosis and treatment of cervical cancer, providing a theoretical basis for clinical diagnosis and treatment of cervical cancer.
