Background and Purpose:Esophageal cancer is a malignant tumor formed by malignant transformation of esophageal epithelial cells.Esophageal squamous cell carcinoma is one of the most common cancers in China,so it has become the focus of research in recent years.With the improvement of medical treatment methods,new treatments for malignant tumors are also being developed.Currently,newly emerged targeted therapeutic drugs for esophageal cancer,such as remiloxacin,are considered to have certain therapeutic effects.However,all the targeted drugs on the market are effective in the treatment of esophageal adenocarcinoma,while no significant effect has been observed in the treatment of esophageal squamous cell carcinoma.At present,there are few reports on targeted drugs for esophageal squamous cell carcinoma,so more scientific research efforts are still needed for the targeted treatment of esophageal squamous cell carcinoma.Vascular endothelial growth factor C(VEGFC)plays a role in promoting the generation of vascular endothelium and lymphatic vessels,while cortical actin(CTTN)is a functional protein that is believed to be related to cell movement.Through previous experiments,we found that the invasion depth,TNM stage,lymph node metastasis and lymphatic infiltration of esophageal squamous cell carcinoma were related to the expressions of VEGFC and CTTN in the esophageal squamous cell carcinoma.However,this experiment is only limited to the study of in vitro tissue,and its results have not been verified by living cell experiment and clinical experiment.The purpose of this study was to investigate the effects of interfering with expression of VEGFC and CTTN on the proliferation and apoptosis of esophageal squamous cell carcinoma,respectively,and to compare the effects of the two genes on the proliferation and apoptosis of esophageal squamous cell carcinoma,so as to lay a certain theoretical foundation for the targeted treatment of esophageal squamous cell carcinoma.Methods:The exexperiment was divided into six groups:blank group?MATE group ? negative control group ? positive control group ? VEGFCsiRNA group ?CTTNsiRNA group.Transient transfection was performed according to experimental groups,VEGFCsiRNA and CTTNsiRNA were successfully transfected into TE1 cells of human esophageal squamous cell carcinoma to interfere with the expression of VEGFCand CTTN.The transfection results were verified by rt-pcr,cell counting kit-8 detects the cell proliferation rate of each group,flow cytometry detects the apoptosis rate.In addition,Statistical software was used to analyze and compare the changes of experimental data in each group.Results : 1.The results of RT-PCR:The m RNA expression of VEGFC in VEGFCsiRNA group and the m RNA expression of CTTN in CTTNsiRNA group were significantly reduced compared to their control group(P<0.05),the transfection was successful.2.The results of Cell Counting Kit-8 and flow cytometry:Compared to their control group,cell proliferation rate was significantly reduced(P<0.05),but apoptosis rate was significantly increased(P<0.05)in VEGFCsiRNA group and CTTNsiRNA group.3.Compared with CTTNsiRNA group,VEGFCsiRNA group had lower cell proliferation rate(P<0.05)and higher apoptosis rate(P<0.05).Conclusions:1.Interfering with the expression of VEGFC and CTTN can inhibit the proliferation of esophageal squamous cell carcinoma and promote the proliferation,thereby inhibiting the growth of esophageal squamous cell carcinoma.2.VEGFC plays a bigger role than CTTN,in the process of proliferation and apoptosis of esophageal squamous cell carcinoma.
