Fractures seriously affect the function of the affected limb and bring huge economic losses.For patients with conservative treatment of fractures or/and Postoperative fracture,there are various methods to promote bone healing.However,there are still 5-10%fracture patients with fracture nonunion or delayed fracture healing.How to detect patients with fracture non-union in early stage and give early effective interventions to reduce the economic loss during fracture recovery and improve the functional rehabilitation level of the affected limb is still one of the hotspots in clinical orthopedic research.The First Part Effects of ShaoYangShengGuFang Prescription to the Distribution of 18F-NaF on Tibial Fracture in SD RatObjective:To explore the application value of 18F-NaF in evaluation of early fracture healing,and the effects of ShaoYangShengGuFang Prescription of rat fracture healing.Methods:72 SD rats were left tibial fracture modeling.The model rats were randomly divided into ShaoYangShengGuFang group and Ossotide group and Physiological saline group.Every group had 24 rats.After 7,15,30,56 days,6 rats were randomly selected from each of the three groups.The PET/CT scan was performed on the fractured ends of tibial fractures,and the uptake of 18F-NaF at the fracture end was calculated.Results:The 18F-NaF uptake of ShaoYangShengGuFang group and Ossotide group reached its peak at15 days?9.00±0.42?8.10±0.71%ID/g?,while the 18F-NaF uptake of Physiological saline group reached its peak at 30 days?4.25±0.35%ID/g?.After 7,15,30,56days,there was no significant difference between the ShaoYangShengGuFang group and the Ossotide group in the 18F-NaF uptake of rat tibial fracture bone?P>0.05?,while the 18F-NaF uptake of the ShaoYangShengGuFang group and the Ossotide group was higher than that of the Physiological saline group,and there was remarkable statistical difference?P<0.05?.Conclusions:18F-NaF has some values in evaluating early fracture healing,and ShaoYangShengGuFang Prescription can promote the healing of tibial fractures in rats.The Second Part Effect of ShaoYangShengGuFang Prescription to the fracture healing in rats and the expression of TGF-?1Objective:To study ShaoYangShengGuFang Prescription on rat fracture healing and the mechanism.Methods:The rats were randomly divided to 3groups:ShaoYangShengGuFang group,Ossotide group and Physiological saline group?n=24?.After 7,15,30,56 days,Every group randomly selected 6rats for X-ray,weighing the wet weight of tibia,testing the serum level of transforming growth factor beta 1?TGF-?1?by enzyme linked immunosorbent assay;andthe expression of TGF-?1inthefractureend was detected by immunohistochemistry.Results:?1?The X-ray:15 days after fixation,there was some vague primary callus formation in every group.30days after fixation,in ShaoYangShengGuFang group and Ossotide group,the number of callus formation in bone stump was significantly higher than that in Physiological saline group,and the density of callus was higher,and the line of fracture was indistinct.56 days after fixation,the fracture healing degree of ShaoYangShengGuFang group and Ossotide group was obviously better than that in Physiological saline group.?2?Tibial wet weight:At four different time points after fracture,the tibial wet weight in ShaoYangShengGuFang group were 0.64±0.05,0.90±0.06,1.03±0.05,1.20±0.05 g;And there was no statistically significant difference compared with Ossotide group?0.67±0.05,0.88±0.06,1.01±0.03,1.18±0.05 g??P>0.05?.While at four different time points,ShaoYangShengGuFang group and Ossotide group were individually compared with the Physiological saline group?P<0.05?,and the wet weight of tibia in ShaoYangShengGuFang group and Ossotide group was significantly higher than that of group C.?3?Serum TGF-?1:7 days after operation,there was no significant difference in serum TGF-?1 levels among the three groups?P>0.05?.15 days after operation,the level of serum TGF-?1 reached the peak value?19.05±0.53ug/L?in ShaoYangShengGuFang group,and gradually decreased to thirtieth days later.At different times,the serum levels of TGF-?1 in ShaoYangShengGuFang group were not significantly different from those in Ossotide group?P>0.05?.while on the 15,30 and 56 days after operation,the levels of TGF-?1 in ShaoYangShengGuFang group and Ossotide group were significantly higher than those in Physiological saline group?P<0.05?.?3?The fracture part of TGF-?1:At four different time points,ShaoYangShengGuFang group and Ossotide group were better than Physiological saline group?P<0.05?.While there was no statistical difference between the ShaoYangShengGuFang group and Ossotide group?P>0.05?.Conclusions:ShaoYangShengGuFang Prescription can improve the expression of TGF-?1 in the rat serum and the fracture end,and promote the formation of callus in the tibial fracture segment,and accelerate bone healing.
