Expression And Significance Of Ciz1-V Protein In Ovarian Serous Adenocarcinoma

Objective:Chemiluminescence was used in our study to analyze the levels of serum tumor markers CA125,CA19-9,AFP and CEA in 167 healthy subjects,136 patients with benign ovarian tumors and 138 patients with ovarian malignancies,analyzing the clinical value of serum tumor markers CA125,CA19-9,AFP and CEA in the diagnosis of ovarian tumors。Immunohistochemistry was used in our study to detect the expression levels of Cizl-V protein in 5 cases of normal fallopian tube specimens,13 cases of benign tumors specimens,9 cases of borderline tumor specimens and 40 cases of ovarian serous adenocarcinoma specimens,analyzing the expression and interaction of Cizl-V protein in these four tissues.We also explored the relationship between the expression of the protein in ovarian serous adenocarcinoma and the patients’clinicopathological features.Meanwhile,prognostic factors associated with survival curves were analyzed to provide a powerful basis for early diagnosis,evaluation and prognosis of ovarian serous carcinoma.Methods:Chemiluminescence and Immunohistochemistry were used in our study and all analyses were performed by SPSS 17.0 software.First of all,we analyzed the levels of serum tumor markers CA125,CA19-9,AFP and CEA in these selected cases.The second,we focused on the relevant research data of Cizl-V protein.The data of the selected cases was counted and further histopathological classification was performed while the expression of Cizl-V protein was detected by immunohistochemistry.We reviewed the clinical data of the selected cases,including age,pathological diagnosis,FIGO staging,TNM staging,tumor classification,date of operation,pelvic lymph node metastasis,recurrence and other related data.We used SPSS 17.0 software to analyze the clinical data and the expression of Cizl-V protein.All measurement data was expressed as mean ± standard deviation(mean ± SD)and results were analysed by two sample independent t-test.The relationship between the expression of Cizl-V protein and clinicopathological features was analyzed by Chi-square test or Fisher exact probability method and the prognostic factors wered screened by Kaplan-Meier single factor survival analysis.Results:Cizl-V protein is mainly localized in cytoplasm and the positive result is brownish yellow.In this experiment,the expression of Cizl-V protein was judged by double scoring method,analyzing the positive rate and staining intensity of colored cells comprehensively and indicating the final score by the form of(-),(+),(+ +),(+ + +).In the present study,the expression of Cizl-V protein in ovarian serous adenocarcinoma was significantly lower than that in non-ovarian serous adenocarcinoma cases(P<0.001).In ovarian serous adenocarcinoma the expression of Cizl-V protein was related with tumor grading(P<0.001)and FIGO staging(P=0.042),but unrelated with age(P = 0.749),TNM staging(P = 0.328)and pelvic lymph node metastasis(P = 0.545).The Kaplan-Meier survival curvesurvival curves showed that patients with Cizl-V protein had a higher survival rate than patients without Cizl-V protein(P = 0.016)and the prognosis of patients with low grade ovarian serous adenocarcinoma was significantly better than patients with high-grade ovarian serous adenocarcinoma(P = 0.035).Conclusion:The expression of Cizl-V protein in normal fallopian tube specimens,benign tumor specimens and borderline tumor specimens showed no difference,but it was higher than that of ovarian serous adenocarcinoma.In normal fallopian tube specimens,benign tumor specimens,borderline tumor specimens and ovarian serous adenocarcinoma specimens,the overall trend of Cizl-V protein expression was gradually lowered.The expression of Cizl-V protein in ovarian serous adenocarcinoma was related to tumor grading and FIGO staging while the prognosis was also related to the expression level of Cizl-V protein and tumor grading,suggesting that Cizl-V protein might be used as a new tumor marker in clinical examination.
