| Objective: This study explored the relationship between RhoA/ROCK1 and bone destruction grade,ISS stage,DS stage,and clinically relevant biochemical indicators by detecting the expression of RhoA/ROCK1 in multiple myeloma(MM)and further understand the role of RhoA/ROCK1 in the development of MM.Methods: The bone marrow specimens of 46 patients with MM who were hospitalized in the Affiliated Hospital of Guilin Medical University and the Second Affiliated Hospital from April to December 2017 were collected as experimental groups,and then divided into mild groups(grade1+2)according to the degree of bone destruction.Severe group(Grade 3)(22males and 24 females,aged 47-75 years,median age 61 years;13 newly diagnosed patients,33 relapsed patients;immunoglobulin typing: IgG-λ 10 cases,13 cases of IgG-k,10 cases of IgA-λ,6 cases of IgA-k,2 cases of IgG,4 cases of λ light chain type,1 case of κ light chain type;DS stage: 4 cases of stage I,6cases of stage II,III 36 cases;ISS staging: 14 cases in stage I,17 cases in stage II,15 cases in stage III;grade of bone destruction: 10 cases of grade 1,6 cases of grade 2,30 cases of grade 3);Then the expression levels of theRhoA-protein and ROCK1-protein in bone marrow supernatant were detected by enzyme-linked immunosorbent assay(ELISA).The expression levels of RhoA and ROCK1 mRNA in bone marrow mononuclear cells were detected by real-time quantitative polymerase chain reaction(qRT-PCR),and the relationship between these data and clinical parameters of MM patients was analyzed.All experimental data were processed with spss20.0,the measurement data were expressed as mean ± standard deviation,the count data and grade data were expressed as median M(min,max),and the mean of the two parameters was compared by t-test or ANOVA.The two-factor correlation analysis measurement data were analyzed by Pearson correlation,and the grade data was analyzed by Spearman.The test level was α=0.05.Results: 1.The age and sex data of the experimental group and the control group were comparable.2.The protein and mRNA expression levels of RhoA and ROCK1 in the experimental group were higher than those in the control group(P<0.05).The protein and mRNA levels of RhoA in grade 3 were higher than grade 1 + 2(P < 0.05).The mRNA expression level of ROCK1 in grade 3 was higher than grade 1+2(P<0.05),and the expression level of protein increased in the two groups,there was no statistical difference(P>0.05).5.RhoA protein expression levels andαCa(r=0.453,p=0.002),bone destruction grade(r=-0.466,p=0.001),β2-mg(r=0.323,p=0.029),ISS stage(r=0.324),p=0.028)and DS stage(r=0.472,p=0.001)showed a positive correlation and negative correlation with Hb(r=-0,696,p=0.000);ROCK1 protein expression level and αCa(r=0.410,p=0.005),bone destruction grade(r=0.332,p=0.024),β2-mg(r=0.331,p=0.025)were positively correlated and negatively correlated with Hb(r=-0.562,p=0.000).Conclusions:(1)RhoA/ROCK1 can reflect the extent of bone destruction;(2)RhoA/ROCK1 can reflect the tumor load level in MM patient;(3)RhoA/ROCK1 may play an important role in the mechanism of MM. |
