OBJECTIVE: Hashimoto's thyroiditis(HT)is a common autoimmune disease,zinc finger protein A20(A20)plays an important role in the negative regulation of inflammation and immunity,and is involved in the pathogenesis of many autoimmune diseases.To discuss the expression of zinc finger protein A20 in peripheral blood of patients with Hashimoto's thyroiditis at the gene level and its relationship with the pathogenesis of Hashimoto's thyroiditis.This study investigated the difference of zinc finger protein A20 mRNA expression in peripheral blood mononuclear cells(PBMC)of patients with Hashimoto's thyroiditis and normal people and the relationship between the expression level of zinc finger protein A20 mRNA in PBMC of HT patients and the clinical indexes of HT: anti-thyroperoxidase antibody(TPOAb)and anti-thyroglobulin antibody(TgAb).Methods: 30 patients with Hashimoto's thyroiditis and 30 healthy people were collected according to the inclusion criteria and exclusion criteria,and got the venous blood of them.PBMC was isolated by Ficoll method,total RNA was extracted with Trizol,and then cDNA was synthesized by reverse transcription.Real-time fluorescence quantitative PCR technique was used to determine the relative expression of zinc finger protein A20 mRNA in PBMC of patients with Hashimoto's thyroiditis,and compared with30 healthy controls in the same period.RESULTS:(1)The expression of A20 mRNA in PBMC of patients with Hashimoto's thyroiditis was significantly lower than that of healthy controls,and the difference was statistically significant(P < 0.001).(2)No correlation is found between the expression level of A20 mRNA in PBMC of Hashimoto's thyroiditis patients and the levels of TPOAb and TgAb in peripheral blood.Conclusion:Zinc finger protein A20 mRNA is low in PBMC of patients with Hashimoto's thyroiditis,zinc finger protein A20 may be involved in the pathogenesis of Hashimoto's thyroiditis.No correlation is found between the expression of A20 mRNA in PBMC andthe levels of TPOAb and TgAb in peripheral blood of HT patients.
