Objective: To investigate the effect of CDKL3 abnormalities on the morphological development of neurite protuberance and the distribution of Golgi,and to explore the mechanism of CDKL3 abnormality on the development of neurite protuberance.Method: The primary cultured hippocampal neurons were randomly divided into CDKL3 knockout group,subtraction rescue group,overexpression group and control group,transfection of neurons in the group of liposomes on the day of culture,train to the desired point,cell immunofluorescence was used,The neuron axons(DIV3,DIV6)or(and)dendrites(DIV6)were labeled in each group,morphological differences of axons and dendrites in each group of neurons under fluorescence confocal microscopy;cell immunofluorescence,the neuron axons(DIV3,DIV6)or(and)dendrites(DIV6)were labeled in each group,at the same time,Golgi bodies were co labeled.The morphological differences of axons and dendrites of neurons and the distribution of Golgi bodies were observed under fluorescence confocal microscope.Result:The number of dendritic branch and length of CDKL3 knockdown group was significantly reduced,and the branch number and length of axons were significantly increased;the number of the group Golgi CDKL3 knock reduction in dendritic distribution of abnormal and remarkably reduce the number of axons in the abnormal distribution of Golgi but a significant increase in the number of.Conclusion: CDKL3 abnormality affects the distribution of Golgi bodies in the neurons,thus affecting the morphological development of the axons and dendrites,and is involved in the pathological mechanism of mental retardation.
