Objective:Through the determination of aging model rats immunoglobulin M(IgM),immunoglobulin A(IgA),immunoglobulin G(IgG),the C3(C3),complement C4(C4)and interleukin 10(IL 10)and interleukin 6(IL-6),interferon-?(IFN-?),tumor necrosis factor-A(TNF-A)the change of the activity,A interferon(IFN-?mRNA)-Y gene,interleukin(IL-10 mRNA)-10 genes of gene expression,as well as the changes of the aging model rats spleen index.In order to provide more experimental evidences for linggui bafa moxibustion with regular moxibustion with open moxibustion to delay senility,it is necessary to study the effect of linggui bafa moxibustion with open moxibustion with open moxibustion on immunological indexes.Methods:forty-three SD rats were randomly divided into A blank group(group A)of 12 rats,and 31 rats were used as model rats.Subacute senescence rat models were prepared by intraperitoneal injection of d-galactose 500mg·kg~-11 once a day for 60 days.After screening,24 rats that had been successfully modeled were divided into model group(group B),and spleen-turtle eight-method treatment group(group C)according to the random number table method,with 12 rats in each group.After the modeling,the blank group and the model group were given no treatment,and the aging rats in the linggui 8 methods treatment group were treated with linggui 8 methods on time for 4 weeks.At the end of the experiment,blood was taken from the main abdominal vein of the rats,and the serum was separated.Rats were sacrificed immediately after blood collection.Spleen tissue was taken from the ice platform and spleen index was calculated by weighing.After weighing,the spleen homogenate was prepared with a manual homogenizer,and the activity changes of C3,C4,IL-10,IL-6,IFN-?and TNF-a in the spleen homogenates of senile rats were determined by enzyme-linked immunoassay.The expression of IFN-?mRNA and IL-10mRNA in spleen homogenate was detected by RT-PCR.Results:(1)The spleen index,serum IgM,IgA,IgG content,spleen tissue C3,C4,IL-10 content and IL-10 mRNA gene expression were greater in the Linggui Bafa treatment group than in the model group significantly;(P<0.05),the difference was significant statistically.(2)The contents of IL-6,TNF-a,IFN-?and IFN-?mRNA in spleen tissues of Linggui Bafa treatment group were lower than those in model group significantly(P<0.05),and increased compared with the blank group significantly(P<0.05).,The difference was significant statistically.Conclusion:(1)Linggui Bafa moxibustion increased the spleen index of aging model rats effectively,increased serum IgM,IgA,IgG,spleen tissue C3,C4,IL-10 levels in aging model rats,and increased The amount of gene expression of IL-10 mRNA.It shows that the eight-method moxibustion of the tortoise promotes the development of the spleen of the immune organs,enhances the immune function and complement system of the body,and effectively regulates the defense function of the body.The eight-method moxibustion of the turtle is subacute caused by D-galactose.The inflammatory reaction plays a certain protective role and achieves the purpose of delaying aging.(2)Linggui Bafa moxibustion reduced the content of IL-6,IFN-?and TNF-a in spleen tissue and decreased the gene expression of IFN-?mRNA.It is indicated that the eight methods of Lingguiregulate the inflammatory factors in the aging model rats effectively,inhibit the immune inflammatory response and regulate the immune balance,and reduce the inflammatory reaction caused by the aging model of D-galactose,and achieve the effect of delaying aging.
