Objective:To investigate the correlation between serum uric acid level and nonalcoholic fatty liver disease in clinical and animal models,and the effect of resveratrol on the inflammation of fatty liver in NAFLD rats with hyperuricemia?Methods:(1)A total of 1,661 patients aged between 20 and 80 years who were diagnosed with NAFLD by abdominal b-ultrasonography,laboratory tests and medical history inquiry in linhai hospital of traditional Chinese medicine and taizhou hospital were selected as subjects;In addition,1727 healthy subjects whose age matched the experimental group were selected as the control group;It was divided into two groups according to male and female gender,and then stratified according to the level of uric acid,and was divided into male and female groups Q1~Q4,and their anthropological indexes and biochemical indexes were compared respectively;Meanwhile,the constitution ratio of TCM constitutions of 80 NAFLD patients with hyperuricemia was analyzed according to the nine-point method of TCM constitutions.(2)48 SPF grade male SD rats were selected and randomly divided into 4 groups after 1week of adaptive feeding : NAFLD group with hyperuricemia(group A),NAFLD group(group B),normal control group(group C),RES treatment group(group D)?Group A was fed with high-yeast and high-fat feed + oxazinic acid emulsion suspension 250mg/kg.d by gavage,group B was fed with high-fat feed,group C was fed with normal feed,and group D was given resveratrol(RES)100mg/kg.d suspension by gavage?The serum biochemical indexes,inflammatory and oxidative factors of rats in each group were measured.Results:(1)Compared with the healthy control group,the body weight,TG,TC,ALT,AST,GGT,FPG,SB,DB,SUA and BMI of both male and female NAFLD groups were increased,while the hdl-c was decreased,also a decrease in age in the male group(P<0.05).Scr was not statistically significant;The levels of age,weight,TG,TC,ldl-c,ALT,AST,GGT,FPG,SB,DB,BMI and Scr in both male and female q4-q1 groups increased with the increase of SUA(P<0.05),while the hdl-c level and the age of the male group decreased with the increase of SUA(P<0.05).(2)The prevalence of NAFLD in male NAFLD groups with different levels of uric acid was 43.2%,47.6%,55.3% and 55.2%(P<0.05),while that in female NAFLD groups with different levels of uric acid was 33.3%,44.3%,58.8% and 77.4%(P<0.05);(3)Pearson correlation analysis showed that body weight,TG,TC,ldl-c,ALT,AST,GGT,SB,DB,BMI and Scr in the male and female groups were positively correlated with SUA(P<0.01),hdl-c was negatively correlated with SUA,and age was negatively correlated with SUA in the male group(P<0.01).(4)Among the 8 skewed constitutions in NAFLD patients with hyperuricemia,the phlegm-wet constitution ratio was the highest(41.25%),followed by qi-deficient constitution(21.25%),accounting for 62.5 of all cases.(5)Animal experiments compared with the normal group,Liver quality was significantly increased in the model group(P<0.05),The NAFLD group also had significantly higher body and liver mass than the NAFLD group(P<0.05).The quality of rat liver in the resveratrol treatment group was lower than that in the model group(P<0.05).The NAS score of the model group and NAFLD group was significantly higher than that of the control group(P<0.01),and the NAS score of the treatment group treated with resveratrol was lower than that of the model group(P<0.01).(6)Model group and NAFLD group of ALT,AST,FFA,SUA,CREA,TNF-a,IL-6,the level of MDA was higher than that of normal control group;SOD and gsh-px levels in both groups were lower than those in the normal control group(P<0.05),The levels of ALT,AST,FFA,UA and il-6 in the model group were higher than those in the NAFLD group(P<0.05).After treatment with resveratrol,the treatment group of ALT,AST,FFA,GLU,UA,CREA,TNF-a,IL-6,the level of MDA was lower,SOD and gsh-px levels in the treatment group were higher than those in the model group(P<0.05).Conclusion:Blood uric acid is closely related to various metabolic indexes.Serum uric acid can aggravate fatty degeneration and inflammation of non-alcoholic fatty liver in inflammatory and oxidative stress pathways,It may be an independent risk factor for NAFLD;Phlegm-dampness and qi deficiency are the main physical types of NAFLD patients with hyperuricemia;Resveratrol can improve liver damage in rats with NAFLD and hyperuricemia?...
