| Objective: To observe the effects of acupuncture method of "Guben peiyuan,Jianpi hewei" on blood lipid levels and liver tissue cells in rats with hyperlipidemia induced by high fat diet.Methods: According to the principle of randomization,40 male SD rats were divided into four groups: blank control group,model group,acupuncture group and Western medicine group,with 10 rats in each group.Rats in blank control group were fed with common diet,while rats in the other three groups were fed with high-fat diet.Four weeks later,blood samples were taken from tail veins of all rats and four items of blood lipids were measured to analyze the model was successful or not.The acupuncture group and the western medicine group were treated for 4 weeks.During the treatment period,the feeds of each group were not changed.The acupuncture group was given acupuncture treatment of "Guben peiyuan,Jianpi hewei" acupuncture every day;Western medicine group Atorvastatin calcium tablets were intragastrically administered;after 4 weeks of intervention,the samples were taken.The contents of TC,TG,LDL-C,HDL-C,etc.in each group were measured,and the structure and morphology of liver tissue cells were observed under an optical microscope.Results:(1)Rat survival status: no abnormal death occurred in the rats in each group during the experiment;the rats in the blank control group had good mental state,free movement,eyes are spirited,strong vigilance,sensitive to external stimuli,Diet and drinking water are normal,The shape,smell and color of the two stools are normal,and the hair is white and shiny.The rats in the model group have poor mental state,multiple lying and less movements,eyes are dull,Vigilance are decline,slow response to external stimuli,eating and drinking are reduced,stool is more sticky,urine smelly,the hair is dull;after 4 weeks of intervention,the physiological state of the acupuncture group and western medicine group(mental state,activity,food intake,urine,hair color,etc.)There was a benign improvement over the model group;and the rats in the acupuncture group had better improvement in stool and hair color than the western medicine group.(2)After the intervention treatment,the body weight and peripheral blood glucose of the acupuncture group decreased compared with the model group,and the difference was significant(P<0.05),which was statistically significant.(3)After intervention,the four levels of blood lipids in the acupuncture group and the western medicine group were significantly improved compared with the model group,the difference was significant(P<0.05),which was statistically significant;the total cholesterol content of the acupuncture group decreased compared with the western medicine.The group was significant(P<0.05),and the difference was statistically significant.The high-density lipoprotein content in the acupuncture group was significantly higher than that in the western medicine group(P<0.05).(4)Observing HE stained liver tissue under 200 times optical microscope,The hepatocytes in the model group were not arranged neatly,and some hepatocytes were enlarged in volume.A large number of vacuolar lipid droplets were observed in the cells;The liver cells of the control group,the acupuncture group and the western medicine group were arranged neatly,the cell size and morphology were basically the same,and a small amount of lipid droplet structure was still observed in the western medicine group.Conclusion:(1)the acupuncture method of "Guben peiyuan,Jianpi hewei" and Western medicine atorvastatin calcium tablets can improve the living state of hyperlipidemia rats to some extent,such as mental state,activity,food intake,urine,hair color,etc.The blood lipid levels of hyperlipidemia rats were significantly reduced.Both were effective in improving the fatty degeneration of rat liver cells and had a benign effect on the structure of liver tissue cells.The acupuncture method was better than Western medicine group in reducing TC and raising HDL.Acupuncture can not only reduce the blood lipid level of rats,but also reduce the weight and blood sugar level of rats.(2)Acupuncture method of “Guben peiyuan,Jianpi hewei”has a significant effect in the treatment of hyperlipidemia and can provide certain experimental data support for clinical application. |
