Zanthoxylum bungeanum Maxim belongs to Rutaceae family and mainly distributed in East Asia and North America and it is widely distributed in many provinces of China.As a Chinese traditional medicinal material contained in the 2015 edition of the“Chinese Pharmacopoeia”,Zanthoxylum bungeanum Maxim has a long history of medicinal use with the main functions of warming middle energizer to relieve pain,killing parasites and relieving itching.In therapeutic practice it has been used to relieve stomachache and bellyache caused by cold,prevent vomiting and stop diarrhea,eliminate enterozoic abdominalgia,and as well as cure eczema and pruritus vulvae in external use.Objective:To study the chemical constituents of Zanthoxylum bungeanum,and to study the antimicrobial activity of the isolated monomer compounds,and to screen the cytotoxicity of the isolated alkaloids,so as to provide promising target for the research and development of new drugs.Methods:The chemical constituents of Zanthoxylum bungeanum stem were isolated and purified by silica gel column chromatography,ODS column chromatography,LH-20 column chromatography,prep-HPLC and recrystallization.The structures of the compounds were identified based on IR,MS and NMR spectra data.Antibacterial experiments were carried out to screen the antimicrobial activities of some extracts and compounds from Zanthoxylum bungeanum.The MTT method was used to evaluate the cytotoxicities of Six alkaloids and?-?-marmesin prepeptone against MC-38?mouse colon cancer cells?,TE-1?human esophageal cancer cells?,MCF-7?human breast cancer cells?,LOVO?human colon cancer cells?,HepG-2?human liver cancer cells?.Results:Twenty-six compounds were isolated from the stems of Zanthoxylum bungeanum and identified as?+?-magnoflorine?1?and dictamine?2?,decarine?3?,zanthobungeanine?4?,pseudocolumbamine?5?,skimmianine?6?,Norchelerythrine?7?,?-?-marmesin?8?,?-?-columbianetin?9?,?-?-decursinol?10?,osthenol?11?,6,7,8-trimethoxycoumarin)dimethylfraxetin?12?,3,4,5-Trimethoxycinnamylalcohol?13?,asarinin?14?,Yangambin?15?,?+?-syringaresinol?16?,ashantin?17?,lupeol?18?,?-amyrin?19?,?-amyrin?20?,?-amyrin?21?,quercetin?22?,rutin?23?,bis?2-ethylhexyl?benzene-1,2-dicarboxylate?24?,24-propylcholesterol?25?,sucrose?26?.The petroleum exacts of the roots and fruits showed strong bacteriostasis activity and the monomers from the sterm showed strong inhibition on Pseudomonas aeruginosa and Bacillus subtilis,and exibited moderate inhibitory activity on Escherichia coli.Seven compounds showed siginificant cytotoxicity against human lung adenocarcinoma A549 with IC50 below 14?M.And Pseudotetrandrine showed broad-spectrum cytotoxicities against six kinds of cancer cell lines.Decarine exibited strong inhibitory activity against HepG-2 with IC50 at 3.75?M.Conclusion:Twenty-six compounds were isolated from the stems of Zanthoxylum bungeanum L.classified to seven alkaloids,ten phenylpropanoids,four triterpenoids,two flavonoids,one benzoic acid,one sterol and one sugar.The extracts of zanthoxylum bungeanum and some compounds from it showed siginificant antibacterial and antitumor activities.The research results provide the basis for further utilization and research of this plant.
