Alcoholic liver disease(ALD)caused by alcoholic liver injury has become the second largest liver disease after viral hepatitis.Therefore,the pathogenesis of ALD and its prevention and treatment have attracted more and more attention.Taking some medicines for a long time will make the body resistant,and it will also have certain side effects.Hence,it seems necessary and significant to explore natural and effective hepatoprotective drugs.Auricularia cornea var.Li.belongs to Basidiomycota,Agaricomycetes,Auriculariales,Auriculariaceae,Auricularia,which is a white mutant strain of A.cornea Ehrenb.In this experiment,A.cornea polysaccharides(APS)and enzymatic-extractable APS(EAPS)were used as the research materials to analyze monosaccharide composition and antioxidant capacity in vitro.Antioxidant,anti-inflammatory and hepatoprotective activities in vivo were measured by acute alcohol-induced liver injury model.The main results are as follows:(1)The results of gas chromatography(GC)analysis showed that APS contained fucose,arabinose,xylose,mannose,galactose and glucose with mass fraction of 9.98%,1.55%,32.7%,11.97%,7.65% and 36.15%,the molar ratio of 5.9:1.0:21.1:6.4:2.3:53.7.EAPS contained fucose,ribose,xylose,mannose,galactose and glucose with the mass fraction of 7.51%,0.78%,20.53%,7.64%,9.33% and 54.21%,the molar ratio of 8.8:1.0:26.4:8.2:10.0:58.1.Fourier transform infrared spectroscopy(FT-IR)analysis showed that both polysaccharides had characteristic absorption peaks of polysaccharides,and all of them were α-type glycosidic.(2)The scavenging hydroxyl,superoxide anion and DPPH radical rate and reducing power of APS and EAPS in vitro were estimated.The results showed that both polysaccharides had certain antioxidant activity,especially EAPS.(3)The APS and EAPS can significantly reduce the activities of aspartate aminotransferase(AST)and alanine transaminase(ALT)in serum,the level of triglyceride(TG)in liver,alleviate lipid metabolism,increase the activities of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and catalase(CAT),enhance the ability of scavenging free radicals generated by alcohol metabolism,inhibit the production of lipid peroxidation product malondialdehyde(MDA),reduce levels of inflammation index nitric oxide synthase inducible nitric oxide synthase(iNOS),cyclooxygenase-2(COX-2),tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and interleukin-1β(IL-1β),elevate alcohol metabolism enzymes of alcohol dehydrogenase(ADH),aldehyde dehydrogenase(ALDH)and cytochrome P4502E1(CYP2E1)content.In addition,histopathological sections of mice liver gradually became normal with the intervention of two polysaccharides.This study suggested that APS and EAPS could improve the antioxidant capacity,improve lipid metabolism,reduce inflammation,relieve alcohol metabolism to reduce the degree of liver tissue damage,and have excellent protective effects on ethanol-induced liver damages.
