ObjectiveIn China,The course of chronic liver disease(CLD)is HBV i nfection – hepatitis – cirrhosis-hepatocarcinoma,generally.Chinese medicine plays an important role in the diagnosis and treatment disease.Moreover,liver-depression and spleen-defic iency syndrome is one of the main syndromes in CLD.Platelets c ontain a large amount of RNA,which plays a non-negligible role in the progression of CLD.This study intends to carry out two researches: 1)TCM Syndrome and Drug Laws of Chinese Medicin e Treatment of CLD by Wu Shoushan Based on Data Mining;2)The Analysis of Chronic Hepatitis B and Liver Cirrhosis Based on p eripheral Platelet RNA-seq,to explore the potential biological characteristics and related signal pathways in course of disea se,as well as the medicine law for treating CLD.MethodsResearch 1: The HIS and Microsoft Excel 2007 were used for data inputting,cleaning,and statistics analyzing.Symptom Features,the frequency of various herbs,association rules analysis and major formula were discussed.Research 2:Individuals were enrolled from the follow-up pat ients of the Institute of Liver Diseases,Hubei Provincial Hos pital of Traditional Chinese Medicine.RNA was extracted aftercollecting and separating the peripheral platelets.Secondly,q uality control of platelet RNA conducted and then sequenced,an d compared with the hg19 reference genome.Differentially expre ssed genes were screened accord with FDR < 0.001 and fold chan ge ? 1.5.Finally,the Bioinformation Analysis conducted in CH B(liver-depression and spleen-deficiency syndrome)-HD,cirrho sis-CHB(liver-depression and spleen-deficiency syndrome).Seque ncing results were analyzed for gene expression level,Gene Ont ology(GO),KEGG pathway and protein-protein interaction(PPI).ResultsResearch 1: Study suggests that 259 prescription data for CHB collected,and the damp-heat accumulation syndrome was the most typical syndrome.The drug frequency and correlation of poria cocos,artemisia argyi,tangerine peel,salvia miltiorrhiza and forsythia are relatively high,and the core prescriptions are mainly added or reduced by Hao qin qingdan decoction and Daan Wan.894 formulations enrolled for the treatment of cirrhosis,the frequency and correlation degree of poria cocos,atractylopathy,licorice root,salvia miltiorrhiza,artemisiae Scopariae were ranked first.The core prescriptions were Jianpi Wan and pinellia pinelliae decoction.Liver-depression and spleen-deficiency syndrome is the most typical syndrome in diagnosis and treatment.Research 2: The general information shows that 131 individuals were enrolled,including 52 HD,39 CHB,40 cirrhosis,and the overall average age was 44.45±3.08.Based on the transcriptome analysis,Transcriptome sequencing results have higher coverage and uniformity.The differentiallyexpressed genes in CHB-HD,cirrhosis-CHB were 144 and 103,respectively.In bioinformatics analysis,for the up-regulated genes in CHB,the highly enriched GO terms are associated with molecular function and RNA interaction/binding,protein-specific interaction/binding and maintenance of ribosome integrity.The biological process of down-regulating differential transcription genes is mainly related to muscle contraction.For the up-regulated genes,the highly enriched GO terms in cirrhosis involved in oxygen transfer,hydrogen peroxide reaction,the down-regulated genes are mainly enriched in biological processes such as epidermal development and ATP synthesis.KEGG analysis showed that up-regulated genes in CHB mainly involved in ribosomal signaling pathway,down-regulated genes mainly involved in cell adhesion,lipid metabolism and amino acid metabolism pathway.most of the genes are enriched into the Hippo signaling pathway in up-regulated genes of cirrhosis and the down-regulated transcriptional genes are mainly enriched in the signaling pathway in oxidative phosphorylation.PPI network analysis indicated that the ribosome family was at the core node in CHB,while the keratin family was at the key node in cirrhosis.Conclusions1.Data mining shows the dampness and heat accumulation syndrome diagnosed by professor wu shoushan as the most typical syndrome in CHB;Liver-depression and spleen-deficiency syndrome is the most typical syndrome of cirrhosis.The syndrome elements of chronic CHB to cirrhosis developed from "toxin" to "toxin,phlegm,stasis and deficiency".Professor Wu Shoushan treats CHB start from damp heat,make good use of clear solution detoxification to benefit wet medicine,treat liver cirrhosis basically to start from recuperating liver and spleen,pay attention to recuperating spleen and stomach,liver and spleen are treated together.2.Study has shown that in the stage of CHB,the ribosome signaling pathway plays a key role in up-regulating the transcriptional genes,suggesting that ribosomes may pass various signals controlling cell growth during the period of disease inflammation or early liver fibrosis.The conduction pathway regulates the cell cycle.The enrichment of the biological process of down-regulating genes in ATP synthesis suggests an abnormal metabolism of energy.Secondly,the significant enrichment of Hippo signaling pathway in cirrhosis suggests hepatocyte apoptosis and aplastic disorder in the progression stage,and further suggests the potential risk of tumorigenesis.In this study,the keratin family members in the stage of cirrhosis need further verified.
