Background: Colorectal polyps are localized uplift arising from the intestinal mucosal epithelium and have the potential to develop into colorectal cancer.Intestinal flora promotes the occurrence and development of colorectal cancer.Previous studies have shown that gut dysbiosis exists in patients with colorectal polyps.Intestinal flora is related to diet,region and other factors.Currently,there are few studies on the characteristics of colorectal polyps patients in guangxi population.Objective:To investigate the difference of intestinal flora between patients with colorectal polyp and those with negative colonoscopy in guangxi.Methods : Fecal samples were collected from patients with colorectal polyps and normal controls who visited the First Affiliated Hospital of Guangxi Medical University from June 2018 to January 2019.The total DNA of fecal samples was extracted with the kit,338 F and 806 R were used as primers to amplify the V4 hypervariable regions of 16 S rDNA gene.The amplification product was purified with gel recovery kit,then purified amplification fragmentlibrary was constructed on the Miseq PE300 platform.High-throughput sequencing was performed on the i-sanger cloud platform and data were analyzed using statistical methods.(Jiangsu,China)from 2014 to 2016.Results:Fecal samples were obtained from 7 patients with colorectal polyps and 46 healthy volunteers.(1)There was no significant difference in fecal bacterial community richness between the colorectal polyps group a nd the normal controls(Shannon Index:P= 0.703).(2)Microbialstructure i n colorectal polyps group issignifcantly different from those in normal con trols.At the phyla level,Firmicutes,Bacteroidetes,and Proteobacteria w ere the main components in both the colorectal polyps and normal control s,and the relative abundance of Fusobacteria in the colorectal polypswas significantly lower than that in the normal controls(P=0.046).(3)At the family level,both the colorectal polyps and normal controls were dominat ed by Bacteroidaceae,Lachnospiraceae,and Ruminococcaceae.Among the m,the relative abundance of Burkholderiaceae(P=0.032),Alcaligenaceae(P=0.034)and Fusobacteriaceae(P=0.039)in the colorectal polyps was si gnificantly lower than that in normal controls.(4)At genus level,Bacteroid es,Faecalibacterium and Lachnospira were dominant in colorectal polyps and normal controls.The relative abundance of Burkholderia-Paraburkhold eria in colorectal polyps was significantly lower than that in normal contr ols(P=0.022).(5)After the normal controls was divided into HC1 group(a ge ?40 years old)and HC2 group(age <40 years old),there was no sign ificant difference in the abundance and diversity of intestinal flora among the three groups(Shannon index: P = 0.661),the relative abu-ndance of Veillonellaceae(P=0.023)and Burkholderia-Paraburkholde-ria(P=0.029)in fecal samples of the three groups was significantly differe-nt.The relative abundance of Veillonellaceae in the colorectal polyps was significantly lower than that of HC1 group(P=0.017)HC2 group(P=0.028),and the relative abundance of Burkholderia-Paraburkholde-ria in the colorectal pol yps was significantly lower than that of HC2 group(P=0.014).Conclusion:This study reveals the characteristics and altered flora of intestinal microbiota in patients with colorectal polyps in guangxi.
