Objective:To determine the effect of RSV-induced DC autophagy on airway remodeling in asthmatic mice,to provide evidence for the role of RSV-induced DC autophagy in the development of asthmatic sputum,clear Wuhu Decoction intervention virus induced DC autophagy induced asthma,a possible link affecting the development of Fuxi,to elucidate the mechanism of Wuhu Decoction in preventing and treating asthma.Methods:160 specific pathogen-free Bal B/C mice,randomly select 30 as the normal group according to the random number table method,the remaining 130 were stimulated with RSV/OVA for 2 weeks,after 2 weeks,ten airway hyperresponsiveness evaluation models were randomly selected from 30 of the normal group and 130 of the model group,the remaining 120 mice in the model group were divided into 6groups according to the random number table,model group,Wuhutang high,medium and low dose groups,positive control group(dexamethasone),rapamycin group(from Induction inducer),20 in each group,Wuhu Tang high,medium and low doses of 20ml/ kg,10 ml / kg,5ml / kg prepared five tiger soup,the positive control group was given dexamethasone,the rapamycin group is given rapamycin,keep the intragastric and intraperitoneal injection solutions consistent in each group,dosing for two weeks,observe lung tissue HE staining,airway MASSON staining,the expression of LC3 I,LC3 II,?-SMA and TGF-? protein was detected by Western blotting.Results:There was significant difference in airway resistance between the model group and the normal group(P<0.05);Pathological manifestations of inflammatory cell infiltration,bronchial mucosal edema,stenosis,subepithelial collagen fiber deposition,smooth muscle thickening,and disordered arrangement were found in the lung and trachea of the model group;The lung tissues of the model group weresignificantly up-regulated(P<0.05)in LC3-I,LC3-II,tracheal tissue ?-SMA and TGF-?.Compared with the model group:In the high,middle and low doses of Wuhu Decoction,the inflammatory cell infiltration was reduced in the bronchial and vascular peripheral areas of the dexamethasone group,the pulmonary interstitial hyperplasia was reduced,and the airway basement membrane thickening and congestion were relieved;The expression of LC3 I protein in lung tissue of Wuhutang low-dose group and positive control group(dexamethasone)was decreased,the difference was statistically significant(P<0.05);The expressions of ?-SMA and TGF-? protein in Wuhutang high,middle and low dose groups and positive control group(dexamethasone)were decreased,and the difference was statistically significant(P<0.05).Conclusion:1.RSV combined with OVA sensitization and stimulation method can successfully prepare autophagy airway remodeling model of mouse asthma dendritic cells;2.Wuhutang high,medium and low doses reduce pulmonary stromal hyperplasia by improving inflammatory cell infiltration in the lung tissue,bronchus and blood vessels;3.Low-dose Wuhu Decoction can improve the autophagy of dendritic cells to a certain extent and reduce the expression of autophagy-related protein LC3I;4.High,medium and low doses of Wuhu Decoction reduce bronchial wall,smooth muscle thickening and subepithelial collagen deposition,reduce airway responsiveness,reduce airway resistance and down-regulate the expression of ?-SMA and TGF-?,thus slowing down and ease the process of airway remodeling;5.Wuhu Decoction may improve autophagy of dendritic cells,inhibit airway remodeling,affect the development of Fuxi,and prevent and treat asthma.
