Objective:To investigate the effect of geniposide combined with emodin on intestinal mucosal barrier damage of rat with systemic inflammatory response syndrome induced by zymosan.Methods:Intestinal mucosal barrier damage model of rats with systemic inflammatory response syndrome induced by intraperitoneal injection of zymosan-paraffin suspension.The model rats were randomly divided into model group,geniposide group(20mg/kg),emodin group(20 mg/kg),geniposide combined with emodin group(geniposide 20 mg/kg+emodin 20 mg/kg),ulinastatin group(2700U/kg).Drugs were given after intraperitoneal injection of zymosan-paraffin suspension immediately and 12 h later.Then,24 h After modeling,The levels of D-lactate(D-LA),diamine oxidase(DAO),endotoxin(ET),tumor necrosis factor-?(TNF-?),interleukin-1?(IL-1?),interleukin-6(IL-6)and interleukin-10(IL-10)in serum were measured,the intestines of rats were taken for histopathological examination.The protein expression of nuclear transcription factor-KB(NF-KB)and Toll-like receptors 4(TLR-4)in the intestinal tissue were determine by immunohistochemistry.rats m the model group had elevated body temperature,decreased white blood cell count,and significantly increased serum TNF-?,IL-1?,IL-10,IL-6,D-LA,DAO,and ET compared to normal control groups(P<UNK>0.01)Results:Body temperature increased,white blood cell count decreased in model rats,the serum levels of D-LA,DAO,ET,TNF-?,IL-1??IL-6 and IL-10 tin model group rat were significantly higher than those in normal control group(P<0.01),the histopathological examination showed that intestinal mucosal epithelial edema,arrangement of mucosal epithelial cells were disordered,mucosal epithelial inflammatory cells were increased,goblet cells were decreased,lamina propria was congestive.The protein expression of NF-KB and TLR-4 in the intestinal tissue were enhanced(P<0.01).Compared with the model group,the levels of TNF-?,IL-1?,IL-6,D-LA,DAO,ET,(P<0.05,P<0.01)in geniposide and emodin group were decreased(P<0.05,P<0.01),the levels of IL-10 in geniposide and emodin group were increased(P<0.05,P<0.01),Histopathological examination showed that pathological damage in intestinal tissue were reduced,The protein expression of NF-KBp65 and TLR-4 in the intestinal tissue were reduced(P<0.05)Conclusion:The results showed that geniposide and emodin could reduce inflammatory reaction,attenuate the intestine mucosal damage induced by zymosan in rats,geniposide and emodin had protective effect on on intestinal mucosal barrier damage of rat with systemic inflammatory response syndrome induced by zymosan,effect of geniposide combind with emodin was better than that of geniposide and emodin alone.The mechanism may be related to reducing the protein expression of NF-KB and TLR-4.
