Objective By observing the notoginseng total saponin on oxygen induced retinal neovascularization in mice and retinal tissue of vascular endothelial growth factor A(VEGF-A),angiogenin 2(Ang2),basic fibroblast growth factor(b FGF),phosphatidyl inositol 3 kinase(PI3K),and serine/threonine kinase(Akt)expression,the effect of exploration notoginseng total saponin on action mechanism of the oxygen induced retinal neovascularization in mice.It provides a new idea and experimental basis for the future treatment in retinopathy of prematurity(ROP)with traditional Chinese medicine.Methods Oxygen-induced retinopathy(OIR)model was established using the International Smith modelling method.60 C57BL/6J neonatal mice were randomly divided into four groups: group A(normal control group),group B(Panax notoginseng saponins group),group C(Ranibizumab group),and group D(OIR group).The normal control group was reared in the normal air environment until the17 th day after birth(P),and the other three groups of newborn mice were reared in the high-concentration oxygen environment of P7-P11.P12,mice in group B were injected with 1?L panax notoginseng saponins,and mice in group C were injected with 1?L ranibizumab.All mice were sacrificed by P17.Retinal tissues were taken for HE staining and the number of nuclei of neovascular endothelial cells with prominent inner boundary membrane was counted.Heart fluorescence perfusion and retinal slice were used to observe the growth of retinal neovascularization in each group.Immunohistochemical staining was used to detect the positive expression of retinal vascular endothelial growth factor in each group.RT-PCR was used to detect m RNA expression of VEGF-A,Ang2,b FGF,PI3 K and Akt.Western blot was used to detect the expression of VEGF-A and PI3 K.Results HE staining results showed that there were a few neovascularization endothelial nuclei protruding the inner boundary membrane in the total saponins group and a large number of neovascularization endothelial nuclei protruding the inner boundary membrane in the OIR group.According to the observation of retinal patella,the phenomenon of retinal vascular contortion and dilation was serious in the OIR group,and the number of non-perfusion areas increased.The phenomenon of vascular contortion and dilation was significantly reduced in the total saponins group and leizumab group,and the number of non-perfusion areas was small.Immunohistochemical results showed that the expression of VEGF was significantly increased in the OIR group compared with the control group,and there was a small amount of VEGF expression in the total saponins group and the leizumab group.RT-PCR showed that the expressions of VEGF-A,Ang2,b FGF,PI3 K and Akt were up-regulated in the OIR group,and significantly down-regulated in the total saponins group and leizumab group.Western blot results showed that there were a small amount of VEGF-A and PI3 K protein expressions in the normal control group,significantly increased VEGF-A and PI3 K protein expressions in the OIR group,and decreased VEGF-A and PI3 K protein expressions in the total saponins group and leizumab group.Conclusion Panax notoginseng saponins can inhibit oxygen-induced retinal neovascularization in mice by regulating the expression of VEGF-A,Ang2,b FGF,PI3 K and Akt.
