Objective To observe the effects of panax notoginseng saponins on retinal neovascularization and long non-coding RNAs?lnc RNAs?in retinal tissue of newborn mice with hyperoxia-induced retinopathy?OIR?,explore its possible mechanism of action,provide the experimental basis for the treatment of retinopathy of prematurity?ROP?in panax notoginseng saponins.Methodology OIR neonatal mouse models were established using the classical Smith modelling method.80 C57BL/6J newborn mice were randomly divided into four groups: A group?normal control group?,B group?OIR group?,C group?Panax ginseng saponins group?,and D group?PBS group?.The normal control group was kept in a normoxic environment until the 17 th day after birth?Postnatal,P?.P7 to P12,group B,group C,and group D were raised at a high oxygen concentration of 75%.P12,In group C mice,1 ?L of panax notoginseng saponin was intravitreally injected into the vitreous cavity of the mouse,In the D group mice,a 1 ?L PBS sterile solution was injected into the vitreous chamber of the eyeball,and it was kept in the normal air until reaching P17.All mice were sacrificed on P17 and retinal tissues were taken.After retinal plating with heart perfusion fluorescein isothiocyanate?FITC-dextran?,the morphological changes of retinal blood vessels in each group were observed;HE staining was used to measure the number of nuclear endothelial cells in the retinal inner boundary membrane;the expression of VEGF was detected by immunohistochemistry;reverse transcription-PCR?RT-PCR?was used to detect m RNA expression changes of Lnc RNA XLOC150636,XLOC150632,XLOC122045,XLOC122042,XLOC100454,and XLOC170009;western blot was used to detect the expression changes of VEGF and FGF2.Results Retinal flatmount showed that OIR group compared with the normal control group,you can see lots of abnormal retinal morphology,retinal neovascularization plexus and non perfusion area,retinal vascular hyperplasia and bending.retinal vascular hyperplasia and bending significantly reduced after Panax notoginseng saponins treatment,non perfusion area also decreased.A large number of curved hyperplastic vascular plexus can still be seen after PBS treatment.HE staining revealed that OIR group and PBS group significantly broke through the inner nuclear membrane of retinal inner limiting membrane.The normal control group and panax notoginseng saponins group only found a small amount of breakthrough through the inner membrane of the inner limiting membrane.The results of immunohistochemistry showed that a small amount of VEGF was expressed in the normal control group,and the expression of VEGF in the OIR group and the PBS group was significantly increased,and the panax notoginseng saponins group was similar to the normal control group.The expression of XLOC150636 and XLOC150632 in the OIR group was significantly up-regulated by RT-PCR and significantly down-regulated in the panax notoginseng saponins group.There was no significant change in the PBS group.XLOC122045,XLOC122042,XLOC100454,and XLOC170009 were significantly down-regulated in the OIR group and significantly up-regulated in the panax notoginseng saponins group.There was no significant change in the PBS group compared with the OIR group.Conclusion Panax notoginseng can regulate the expression of Lnc RNAs in the retina of OIR model neonatal mice and inhibit the proliferation of retinal neovascularization.
