Object:Using liquid chromatography-mass spectrometry to screen out a variety of differential proteins from renal cell carcinoma,select SERPINA1,and then explore the expression of SERPINA1 in various types of renal cell carcinoma(mainly renal clear cell carcinoma)and its clinical significance.Methods:1.Liquid-mass spectrometry technology to construct differential protein expression profiles of renal cancer:(1)Collect kidney cancer specimens.These kidney cancer specimens must be kidney cancer specimens that have been diagnosed by pathology and excised during surgery.The specimens were prepared into peptides mixture.(2)The protein in the above samples was detected by liquid chromatographymass spectrometry using liquid chromatography and detection system as the separation system to construct differential protein expression profiles of renal cancer;and construct differential protein expression profiles of renal cancer;(3)Screening for a variety of highly expressed proteins in renal cell carcinoma,and reviewing related data,select SERPINA1 as the target protein for research and elaboration.Thirty-six patients with renal cell carcinoma and corresponding paracancerous tissue specimens were removed from the operation.These renal cancer patients did not receive any treatment such as radiotherapy or chemotherapy before surgery.2.Confirmation of the expression of SERPINA1 in the corresponding tissues by Western Blot;3.Immunohistochemical method to verify the expression of SERPINA1 in renal cell carcinoma: patients with clear pathological diagnosis of clear cell renal cell carcinoma,take the cancer tissue section,preserved by paraffin seal,using immunohistochemical experimental methods to detect the expression of SERPINA1 in the diseased group(positive group)and the control group(negative group).After staining,the diseased group and the control group were observed under the microscope,and the image was taken under the microscope to confirm the expression of SERPINA1,and to further understand the relationship between SERPINA1 and clinical pathology of renal cancer patients.Results:1.By liquid chromatography-mass spectrometry,we obtained 41 differential proteins in renal cell carcinoma,of which 36 proteins were up-regulated and 5 proteins were down-regulated.Our target protein SERPINA1 is one of the up-regulated proteins.2.By immunoblotting experiments,it can be proved that the expression level of SERPINA1 is significantly increased in renal cell carcinoma compared with adjacent tissues.3.In the immunohistochemistry experiments of 11 patients with renal cell carcinoma we selected,the tissue sections of patients with renal cell carcinoma were significantly expressed compared with the negative control after staining.Conclusions:1.The expression level of SERPINA1 in renal cell carcinoma is significantly higher than that in adjacent tissues.2.SERPINA1 is highly expressed in cancer tissues of patients with renal cancer.
