Objective To detect the expression of Survivin protein and VEGF protein in the tissue of normal kidney and renal cell carcinoma (RCC) (including different pathological grades and clinical stages). To analyze the correlation of Survivin protein and VEGF protein. To explore the roles of Survivin and VEGF protein in the early and middle development stages of RCC. To discuss the clinical significance of Survivin and VEGF that appears in diagnosis, treatment and prognosis evaluation of RCC. To provide a new way of early-diagnosis and targeted therapy to RCC. Method The expression of Survivin and VEGF protein in the tissue of 32 cases of RCC and 10 cases of normal kidney was examined by Streptavidin-Peroxidase (SP) immunohistochemistry and Western-blot. According to different pathological variables, the RCC patients were divided into different groups and the results were analyzed by SPSS 11.5 for Windows software. Results 1.There was no Survivin and VEGF protein expressed in the tissue of normal kidney.2. In the group with the age of >60 and≦60, Survivin was differently expressed in the rate of 57.9% and 46.2% while in the group with the tumor diameter of >7cm and≦7cm, the expression rate turned out to be 50.0% and 54.5%.The result suggested that there was no statistically significant difference between the two.(p=0.513, 0.811>0.05).3. Survivin protein was significantly expressed in the tissue of RCC with the rate of 53.1%. In the poorly differentiated group, the differentiation group and the well-differentiated group, Survivin was differently expressed in the rate of 100%,66.6%,38.1%.InⅠandⅡperiod group, it was expressed in the rate of 33.3% while inⅢandⅣperiod group, the rate was 90.9%. There was statistically significant difference.4, There was no correlation between the expression rate of VEGF and the age and the size of the tumor of the patients. In the age group (>60 and≦60), Survivin was differently expressed in the rate of 42.1%and 23.1% while in the group with the tumor diameter >7cm and≦7cm, the expression rate changed to 40.0% and 31.8%.The result suggested that there was no statistically significant difference between the two.(p=0.259, 0.653>0.05)5. VEGF protein was significantly expressed in the tissue of RCC with the rate of 34.4%. In the poorly differentiated group, the differentiation group and the well-differentiated group, Survivin was differently expressed in the rate of 100%,33.3% and 19.9%.InⅠandⅡperiod group, VEGF was expressed in the rate of 14.3% while inⅢandⅣperiod group, the rate was72.7%.There was statistically significant difference. 6. A product that concluding the molecular weight of 16.5KD and 21KD was detected with the western-blot in the tissue of RCC while it can't be found in the tissue of normal kidney. 7. The expression rate of Survivin and VEGF is positively correlated in RCC. (r=0.416 P<0.05).Conclusion 1,Respectively, the result of the over- expression and no-expression of Survivin and VEGF in the RCC group and the normal kidney group suggested that they might participate in the development of RCC. 2,The expression intensity of Survivin and VEGF is not relevant to the patients'age and tumor size.3, The expression of Survivin and VEGF in RCC were positively correlated to the pathological grades and clinical stages. The detecting of the two suggests that they might play an important role in the process of diagnosis, treatment and prognosis evaluation of RCC.
