Objective The rat models of trigeminal neuralgia were established by chronic constriction of the left infraorbital nerve(ION-CCI).Injection of Panx 1 specific si RNA was used to block the expression of Pannexin-1(Panx 1)in the trigeminal ganglion.Von Frey hairs,Western blot and q RT-PCR were used to measure the expression of Panx 1 in the trigeminal ganglion and its role in trigeminal neuralgia.Methods The rat models of trigeminal neuralgia(ION-CCI group)was established in male SD rats by chronic constriction of the left infraorbital nerve(ION-CCI),and a sham operation group was also established which only be exposed the infraorbital nerve but not ligated.The mechanical stimulation thresholds of the left whisker pads of the two groups of rats were tested with Von Frey hairs to verify whether the rat model of trigeminal neuralgia was successfully established.ION-CCI rats were randomly divided into two groups: CCI+Panx1-si RNA group which Panx 1 specific si RNA was injected into the trigeminal ganglion region through the left infraorbital foramen of the ION-CCI rat;CCI+Ctl-si RNA Group which negative control si RNA was injected into the trigeminal ganglion region through the left infraorbital foramen of the ION-CCI rat.The expression of Panx 1 m RNA in the trigeminal ganglion of the two groups was compared by q RT-PCR.The expression of Panx 1 protein in the trigeminal ganglion of the two groups was compared by Western blot.The mechanical stimulation pain thresholds of the two groups were compared by Von Frey hairs behavioral test.Results 1.Von Frey hair behavioral test showed that the threshold of mechanical stimulation pain in the area of the tentacles of the ION-CCI rats was significantly lower than that of the sham operation group,indicating that the rat model of trigeminal neuralgia was successfully constructed.2.The results of q RT-PCR and Western blot showed that the expression of Panx 1 in the trigeminal ganglion of the CCI + Panx 1-si RNA group was significantly lower than that of the CCI + Ctl-si RNA group.These results was shown that Panx 1 specific si RNA effectively inhibited the expression of Panx 1 in the trigeminal ganglion.3.Von Frey hair behavior test indicated that the mechanical stimulation threshold of rats in CCI + Panx1-si RNA group was significantly higher than that in CCI + Ctl-si RNA group.Conclusion The rat models of trigeminal neuralgia are established by chronic compression injury of the infraorbital nerve.Inhibition of Panx 1 expression in the trigeminal ganglion can alleviate the pain hypersensitivity response of trigeminal neuralgia.
