Objective To investigate the inhibitory effect of miR-155 on high glucose-induced apoptosis in human lens epithelial cells(HLEpi C)and SD rats.Methods The optimal drug concentration of miR-155 and the effect of different concentrations of glucose medium on cell proliferation were detected by cell MTT assay.The experiment was carried out in vitro.The animal experiment group included: 5.5mol / L normal glucose group(LG),25 mol / L mannitol group(M),25 mol / L high glucose medium group(HG),high sugar medium treated miR-155(30 nmol / L)administration group.After determining the optimal drug concentration,a cell experimental group was established,and HLEpi C was treated with a normal glucose group(LG)to a density of 30%-40%,and different culture groups were set for drug treatment.The culture group consisted of normal glucose group(LG),25 mol / L mannitol group(M),25 mol / L high glucose medium group(HG),high glucose medium treated with 30 nmol / L miR-155 agonist mimic and Drug control CTm group.Western blot was used to detect the difference of the expression of Bax,Bcl-2,caspase-3 and procaspase-9 in different treatment groups.The effect of mimic on HLEpi C cell migration was observed by cell scratch assay.Results Animal experiments showed that when the mimic concentration of miR-155 agonist was 30 nmol / L,the change in lens turbidity caused by high glucose could be changed.The results of cell experiments showed that with the increase of glucose concentration in the medium,the higher the apoptosis-promoting rate of cells,when 30 nmol / L mimic acted on the cells,it had a significant effect on the proliferation and migration of HLEpi C.In western blot,mimic significantly inhibited high glucose-induced apoptosis of HLEpi C and apoptosis of lens proteins in SD rats,and altered apoptosis-related proteins Bax,Bcl-2,caspase-3 and caspase--The expression level of the precursor protein procaspase-9 of 9 was significantly lower than that of the high glucose control group.Conclusion miR-155 can inhibit the apoptosis of HLEpi C and SD rats induced by high glucose.
