Objective:To observe the effect of acupuncture on the content and behavior of Cholecystokinin(CCK),P in serum and brain tissue of PD rats with insomnia.On this basis,the related mechanism of this acupuncture was discussed by comparing it with western medicine and routine acupuncture.Methods:PD model rats were prepared by single-side striatum double-target injection,and the behavior changes induced by APO at the sixth week after surgery were used to verify the success of PD model.On the basis of PD model,PCPA suspension was intraperitoneally injected six weeks later to prepare PD rats with insomnia.At the same time,the normal group,the sham operation group,the model group,the drug group,the routine acupuncture group and the tiaoshenchangqing acupuncture group were set up.The behavioral changes of rats were observed after different intervention manipulations,and brain and blood were taken at the corresponding time points.The effects of CCK and SP in brain tissue and serum were detected by ELISA.Results:(1)Compared with the normal group,the CCK content in serum and brain tissue of the model group was significantly decreased,and the SP content was significantly increased(P<0.01),and the relevant behavioral indexes were significantly different(P<0.05).(2)Compared with the model group,the levels of CCK and SP in the serum and brain tissues of the rats in the drug group,the conventional acupuncture group and the tiaoshenchangqing acupuncture group increased after treatment,while the levels of SP decreased significantly(P<0.05),and the relevant behavioral indicators were significantly different(P<0.05).(3)Compared with the drug group,the tiaoshenchangqing acupuncture group had more significant effects in improving the content of CCK and reducing the content of SP(P<0.05),while the conventional acupuncture group had no significant differences(P>0.05).There was no significant difference in behavioral indicators between the tiaoshenchangqing acupuncture group(P>0.05)and the conventional acupuncture group(P<0.05).(4)Compared with the conventional acupuncture group,the CCK content and SP content in serum and brain tissues of the tiaoshenchangqing acupuncture group were significantly increased(P<0.05).Behavioral indexes were significantly different from those in the conventional acupuncture group(P<0.05).Conclusion:(1)In improving the behavioral symptoms of PD rats with insomnia,the acupuncture method of "tiaoshenchangqing sanliujiu" can achieve the therapeutic effect of western medicine,and it is superior to the conventional acupuncture group.(2)The contents of CCK,SP in serum and brain tissue of PD model rats with insomnia were regulated by acupuncture,routine acupuncture and drug therapy,and the acupuncture method of "regulating mind and feeling" had the best regulating effect,followed by western medicine and routine acupuncture treatment again,and the content of CCK,SP in serum and brain tissue of rats with insomnia was regulated by the acupuncture method of"tiaoshenchangqing sanliujiu",followed by the treatment of western medicine and the treatment of routine acupuncture.(3)One of the mechanisms of "tiaoshenchangqing sanliujiu" acupuncture is to regulate the content of CCK,SP in serum and brain tissue.
