Fructus Sophorae is the fruit of Sophora japonica L..which is a kind of arbor belonging to Leguminosae.It is a common traditional Chinese medicine included in CHP(Chinese Pharmacopoeia)for a long time.Fructus Sophorae have a large amount of flavonoids,which have many pharmacological functions such as lowering blood pressure and lipids,softening blood vessels,anti-oxidation,antibacterial,lowering blood glucose,ect.On the basis of previous studies,the extraction and content determination of Fructus Sophorae,the seeds of Fructus Sophorae and and the callus of seeds were carried out.At the same time,the extraction conditions were optimized.And the inhibitory effects and types of a-glucosidase on the activity of a-glucosidase of them were further studied.The results provide a theoretical basis for the hypoglycemic effect of Fructus Sophorae,Fructus Sophorae seed embryo and embryo callus,and the development of Fructus Sophorae produts can increase the diversification of the products of Fructus Sophorae in the market and meet people's pursuit of health,which has great practical value.The main contents and results are as follows:(1)The content of flavonoids in the extracts from Fructus Sophorae,seed embryo of Fructus Sophorae and the callus of seed embryo of Fructus Sophorae were(55.71±0.96)mg/g,(46.72±0.80)mg/g and(10.78±0.75)mg/g respectively,and the content of flavonoids in Fructus Sophorae was the highest.Ultrasonic assisted ethanol extraction was optimized by orthogonal test.The optimum extraction conditions were as follows:ratio of material to liquid was 1:50,ultrasonic time was 35 min,the ethanol content is 70%.(2)The flavonoids were extracted and determined by ultrasonic assisted ethanol extraction,hot water extraction and cold water extraction.The results showed that the flavonoids contents were(56.36±0.96)mg/g,(52.31±1.33)mg/g and(47.13±0.71)mg/g,respectively.After the difference analysis,it was found that ultrasonic assisted ethanol extraction was significantly better than water extraction,but there was no significant difference between water extraction,so cold water extraction can be chosen in the production of related products.(3)The flavonoids extractive of Fructus Sophorae(IC50 ? ? =22.82mg/L),embryo(IC50embryo=32.60mg/L)and callus(IC50 callus=35.81 mg/L)showed a strong inhibitory activity to against?-glucosidase which wree higher than that of Acarbose(IC50acarbose=905.29mg/L).All of them were shown noncompetitive type model on a-glucosidase and Fructus Sophorae was best.By analyzing the Lineweaver-Burk,the inhibitory types of flavonoid extracts from them on a-glucosidase were all non-competitive.(4)Prepare the product of Fructus Sophorae flavonoids by orthogonal test and optimize the beverage formula with fuzzy mathematical sensory evaluation method.The best beverage formula is AiB2C1D3,that is,Flavonoids crude extract 1.5g,?-cyclodextrin 6g and xylosiclg,and 1g product blended with 30mL hot water.At the same time,through the inhibition of a-glucosidase activity,it was found that compared with the a-glucosidase inhibitor,Acarbose,the product could not achieve the inhibitory effect on a-glucosidase activity.Therefore,the product only contains flavonoids and has a certain auxiliary hypoglycemic effect and can not achieve the therapeutic effect.(5)It was determined that the water content in the product was less than 5%,which was in accordance with the national standard.The ash content is 2.53%,the loss rate of flavonoids is 1.1%,and the content of flavonoids decreases with the increase of water content during 90 days of sealed storage,and the loss of flavonoids is about 2.1%.The moisture content in the storage process was always less than 5%,so the product could be sealed at 25 0C at room temperature.
