Radix sophorae flavescentis of the dry legumes plant's root is a kind of well-known Chinese herbal medicine. Radix sophorae flavescentis have many indications such as cold character, bitter taste , reducing heat, drying wet, insecticidal and diuretic etc.The flavonoids in Radix sophorae flavescentis are very rich and have high medical values and physiological effects. In this paper research we have studied the best extraction condition, antioxidant activity of Flavonoids in Radix sophorae flavescentis ,and analysed primary the flavonoids by Performance Liquid Chromatography- Electrospray/Mass Spectrometry (HPLC-MS) .It will provide a theoretical basis for the application of flavonoids in Radix sophorae flavescentis. The paper is composed of four parts, its primary coverage was as following:The foreword introduces the flavonoids chemical composition in Radix sophorae flavescentis, the biological activity, the isolation and analysis method , as well as the research significance.In the first part the extraction conditions of flavonoids in Radix sophorae flavescentis by supercritical fluid extraction (SFE) and microwave were studied. The results showed that supercritical extraction method was superior than microwave extraction. The effects on the extraction efficiencies of SFE-CO2 extracting temperature, pressure , time and the Ethanol velocity was investigated by both single-factor and orthogonal design. The best extraction conditions were summarized that : extraction temperature was 40℃,extraction pressure was 30 MPa, Ethanol velocity was 0.4 mL/min, extraction time was 60 min. According to the conditions, the extraction efficiency of the flavoniods is as high as 6.44%.In the second part the expiration of the anti-oxidability and the hydroxyl free radical by flavonoids in Radix sophorae flavescentis By the spectrophotometric method and trihydroxybenzene autoidation were studied. The experimental results indicated that: flavonoids in Radix sophorae flavescentis have the high antioxidant activity.In the third part the separating condition of flavonoids of HPLC-MS was determined. The method of HPLC-MS has separated flavonoids in Radix sophorae flavescentis effectively. The HPLC-MS spectrogram will provides the basis to study flavonoids in Radix sophorae flavescentis farther.As one kind of new green isolation techniques, SFE-CO2 has the superiority on extracting of flavonoids in Radix sophorae flavescentis. Flavonoid has the high antioxidant activity, it will be applied in food, the drugs, the cosmetics as one kind of health care ingredient . HPLC-MS may determine qualitatively the component of flavonoid of extraction and provides certain theory basis for the flavonoids in Radix sophorae flavescentis further.
