Objective: To explore the effect of curcumol on proliferation,migration,autophagy and apoptosis in ectopic endometrial stromal cells(EESCs)in adenomyosis in vitro,providing theoretical foundation for clinic treatment of adenomyosis.Methods: Seperated,purified and cultured EESCs in adenomyosis were treated with curcumol in different concentrations,10?l/ml anhydrous alcohol was added in the control group,the cell proliferation rates were detected with CCK-8 method;the cell migration rates were detected with scratch assay;the cell autophagy rates were detected with immunofluorescence technique;and the cell apoptosis rates were detected with Annexin V-FITC/PI flow cytomeutry.Result: 1)The cell proliferation was inhibited in dose-dependent and time-dependent manners after treated with curcumol in different concentrations.2)Positive spots in the cell increased with the increase of drug concentration under the fluorescence microscope,which indicated that curcumol promoted autophagy of EESCs;and the effect had obviously positive correlation with drug concentration;3)The results of flow cytomeutry showed curcumol which the concentration over 25.00?g/ml could induce cell apoptosis after affecting on EESCs for 48 hours;and compared with 25.00?g/ml group,the cell apoptosis rates in 100.00?g/ml group was significantly higher.4)Scratch assay showed that curcumol could inhibit migration of EESCs;and the 100.00?g/ml group had the best function of inhibiting.Conclusion: By inhibiting the proliferation and migration,and inducing cells apoptosis and autophagy in EESCs in adenomyosis in vitro,curcumol has potential therapeutic effects in adenomyosis.
