Objective: To investigate the correlation between the expression of PTEN and the mechanism and clinical characteristics of systemic lupus erythematosus by detecting the expression of PTEN and characteristic cytokines,which were expressioned in mononuclear cells of patients with systemic lupus erythematosus.Methods: 22 systemic lupus erythematosus patients and 20 healthy control were chosen to attend the research according to the inclusion and exclusion criteria.The level of PTEN was assayed by RT-q PCR,and the expression of IL-4?IL-10?IL-17?IFN-? were assayed by ELISA.Comparative and correlation analysis was conducted by SPSS 22.0.Results:1 Expression of PTEN m RNA in peripheral blood PBMCThe relative expression level of PTEN m RNA in PBMC of the experimental group before treatment was significantly lower than the control group and the healthy control group(P<0.05).The relative expression level of PTEN m RNA in the experimental group compared with the healthy control group was not significant difference(P>0.05).2 Correlation between the expression level of PTEN m RNA and disease activityin patients with SLEThe expression of PTEN m RNA in PBMC of SLE patients was negatively correlated with SLEDAI score and anti-ds-DNA antibody,and positively correlated with complement C3(P<0.05),and had no correlation with complement C4 antibody(P> 0.05).The expression level of PTEN m RNA in PBMC of SLE patients in the SLE clinical mild activity group,moderate activity group and severe activity group was statistically significant(P<0.05),and the expression level of PTEN m RNA was negatively correlated with the clinical activity of SLE.The expression level of PTEN m RNA in peripheral blood PBMC of patients with SLE with lupus nephritis or vasculitis was lower than the of patients without lupus nephritis or vasculitis(P<0.05);the expression level of PTEN m RNA in SLE patients with arthritis was no significant difference compared with the expression level of arthritis-free patients(P>0.05).3 The expression of IL-4,IL-10,IL-17 and IFN-? in serumThe expression level of inflammatory factors in serum of patients with SLE before treatment in the experimental group was compared with that after the treatment in the experimental group,the expressions of IL-10 and IFN-? were decreased(P<0.05),and the expression of IL-17 was increased(P<0.05),there was no significant difference in IL-4 expression(P>0.05).The experimental group was compared with the healthy control group before treatment,the expressions of IL-10 and IFN-? were decreased(P<0.05),and the expressions of IL-4 and IL-17 were increased(P<0.05).The experimental group was compared with the healthy control group after treatment,IL-4 expression was increased(P<0.05),IL-10 expression was decreased(P<0.05),and IL-17 and IFN-? expression were not significantly different in the experimental group(P>0.05).0.05).4 Correlation analysis between PTEN m RNA expression and IL-4,IL-10,IL-17 and IFN-?The expression of PTEN m RNA in peripheral blood of SLE patients was negatively correlated with IL-4 and IL-17 levels in serum(P<0.05),and positively correlated with IL-10 and IFN-? expression levels(P<0.05).Conclusions:(1)PTEN may participate in the occurrence and development of SLE by regulating Th1/Th2,Th17/Treg cell axis,and it is suggested that PTEN has the potential to become a potential target for therapy.(2)The expression level of PTEN reflects the disease activity of SLE,and it may become one of the indicators of SLE disease activity.
