Expression And Significance Of HIF-1?, Cyclin E And Cyclin D1 Proteins In Human Colon Cancer Tissues

OBJECTIVE:To observe the changes in the expression of hypoxia inducible factor 1 alpha(HIF-1 alpha),Cyclin E(CyclinE)and Cyclin D1(CyclinD1)in the cancer tissues of patients with colon cancer,and to explore the clinical significance.METHODS:62 cases of colorectal cancer were collected from May 2013 to December 2017 of the First Affiliated Hospital of Guangxi Medical University,with surgical excision and enteroscope biopsy,each of which had colon cancer tissue and its matched normal colonic mucosa.which were included for the first operation,and the immunohistochemical SP method was used to detect colon cancer and normal.With the Cyclin E,Cyclin D1 and HIF-1alpha in colon mucosa analyzed,The expression results of three indexes were analyzed with the clinicopathological features of colon cancer patients.Finally,the correlation analysis between the three expressions was carried out by using the Spearman related method.RESULTS: the positive expression rates of Cyclin E in normal colonic and colon cancer tissues were25.80%(16/62),64.74%(42/62),the HIF-1 alpha22.58%(14/62),72.58%(45/62)and the CyclinD1 22.58%(14/62),72.58%(45/62).The positive expression rate of HIF-1 alpha,Cyclin D1 and Cyclin E in colon normal mucosa and colon cancer tissues was found.(2)The difference was statistically significant(all three P < 0.05).and the same results were related to the list as followed : tumor location,the degree of differentiation,Dukes staging,TNM clinical stage,lymp node metastasis and distant metastasis(P < 0.05).(3)The expression of HIF-1 alpha and Cyclin E in colon cancer tissues was positively correlated(r = 0.272,P < 0.05),and the same as CyclinD1 and HIF-1 alpha(r = 0.276,P < 0.05).as the Cyclin D1 and Cyclin E(r= 0.32,P < 0.05).CONCLUSION:(1)HIF-1 alpha,Cyclin E and Cyclin D1 are highly expressed in colon cancer tissues,and the three may be involved in the development and progression of colon cancer;(2)In colon cancer tissue,the high expression of three Indicators may be related to the proliferation,invasion and metastasis of colon cancer.(3)HIF-1 alpha can promote the high expression of Cyclin D1 and Cyclin E in colorectal cancer cells,and the three Indicators may promote each other in the development of colorectal cancer.
