Objective : To investigate the effect of focal cerebral ischemia-reperfusion injury on the expression of ?-SMA(?-smooth muscle actin)in mouse brain smooth muscle cells.Methods :The experiment used C57BL/6 mice,based on the rat cerebral ischemia model established by the Japanese scholar Koizumi,using the prepared wire plug to embolize the middle cerebral artery(MCA)of the mouse to cause the middle cerebral artery.The corresponding brain tissue ischemic infarction was supplied to prepare a mouse model of focal cerebral ischemia(MCAO model)using Longa behavioral biology score,q RT-PCR,Western blot and immunohistochemistry.Methods: To compare the brain histomorphology and ?-SMA m RNA phase in the middle cerebral artery blood supply area of mice with cerebral ischemia-reperfusion group after cerebral ischemia for 2 h and reperfusion for 24 h(experimental group),sham operation group and blank control group.Corresponding gene expression and?-SMA protein expression and changes in the number of?-SMA positive cells.Results :The results of TTC staining showed that the cerebral infarct size of the experimental group was significantly larger than that of the sham operation group and the blank control group.The striatum and cortex of the experimental group showed a pale ischemic infarction,and The side is edematous and the shape is asymmetrical.The brain tissue of the sham operation group and the blank control group were almost all stained into red,the local texts were clear,and the left and right forms were symmetrical.The results of q RT-PCR showed that the expression of ?-SMA gene in the peripheral region of cerebral infarction was significantly higher than that in the sham operation group and the blank control group after focal cerebral ischemia reperfusion in mice for 2 h and reperfusion for 24 h.(P<0.05).Western blot analysis showed that the expression of ?-SMA protein in the peripheral region of the infarcted area was significantly higher than that in the sham-operated group and the blank control group after focal cerebral ischemia-reperfusion in mice for 2 h and reperfusion for 24 h.Academic significance(P<0.05).Immunohistochemistry showed that the number of ?-SMA positive cells in the peripheral area of ??cerebral infarction increased after cerebral ischemia and reperfusion for 24 h,and the distribution was uneven and the morphology was irregular.The number of?-SMA positive cells in the corresponding areas of the sham operation group and the blank control group was small.Conclusion : Focal cerebral ischemia-reperfusion injury increased the expression of ?-SMA(?-smooth muscle actin)m RNA and protein in mouse vascular smooth muscle cells,and the ?-SMA positive cells in the peripheral area of cerebral infarction increased significantly,suggesting cerebral vascular smooth muscle.Cells play an important role in focal cerebral ischemia-reperfusion injury.
