Background:The aim of the study was to investigate the prognostic role of mannose receptor expression and the interaction with its ligand mannose in gastric cancer patients.Methods:NPL?Narcissus pseudonarcissus lectin?lectin histochemical staining and immunohistochemical envision two-step assays were carried out to detect the expression of NPL and MR in 50 cases of primary advanced gastric adenocarcinoma tissues from the Gastrointestinal Surgery of the First Affiliated Hospital of Anhui Medical University.Additionally,10 adjacent tumor tissues were selected as healthy controls.The relationships of NPL and MR expression with clinicopathological features of gastric cancer patients were analyzed.Results:?1?In gastric cancer tissues,NPL positive staining was mainly localized to the cytoplasm or envelope of the cancer cells,and the expression of NPL was significantly high than in normal tissues adjacent to the cancer?p=0.003?.In gastric cancer stromal tissues,NPL positive staining was located in the cytoplasm or envelope of macrophages.The mean density of NPL+macrophages in gastric cancer tissues was29.32±13.76/HPF,while the density of NPL+macrophages in normal tissues adjacent to the cancer was only 20.14±7.41/HPF.The density of NPL+macrophages in gastric cancer interstitial was significantly higher than that in adjacent tissues,and the difference was statistically significant?p=0.006?;?2?The expression of NPL in gastric cancer tissues was significantly correlated with TNM stage of patients?p=0.02?but was not correlated with other clinicopathological parameters.There was no correlation between NPL+macrophages in gastric cancer stromal and clinical pathological characteristics of patients;?3?MR staining was localized to the cytoplasm or envelope of macrophages in the tumor stromal and the mean MR+macrophages density was20.69±15.52/HPF.The density of MR+macrophages in normal tissues adjacent to cancer was only 7.56±2.86/HPF.The MR positive density in gastric cancer stromal tissues was significantly higher than that in adjacent normal tissues,and the difference was statistically significant?p<0.001?;?4?The MR+macrophages was significantly associated with TNM staging and histological typing?p=0.009,p=0.04?,but was no correlation with clinical pathological parameters such as age,sex,tumor size,location,depth of low postive macrophages?Log-rank time series test,p=0.037?.And the patients with high density of MR+macrophages had significantly lower postoperative survival than those with low postive regression univariate analysis found that tumor size,lymph node metastasis,distant metastasis,TNM staging,and NPL+macrophages and MR+macrophages in gastric cancer stromal tissues were significantly associated with patient prognosis.Multivariate analysis revealed that lymph node metastasis and MR+macrophages in gastric cancer stromal were independent prognostic factors for gastric cancer patients,but NPL was not.Conclusion:?1?Both the molecules NPL and MR in gastric cancer tissues was highly expressed?p<0.05?,which might indicate that the high expression of mannose and MR was involved in the occurrence and invasiveness of gastric cancer;?2?The expression of NPL and MR in gastric cancer tissues was related to the clinicopathological and biological characteristics of gastric cancer.NPL and MR expression were among the important molecular biological markers evaluating the biological characteristics of gastric cancer;?3?There was a significant negative correlation between NPL and MR expression in gastric cancer stromal tissues?p=0.009,r=-0.363?.Mannose might affect the progression of gastric cancer by regulating MR expression;?4?This study demonstrated that tumor of NPL+/MR+macrophages in gastric cancer stromal revealed that OS in the lowNPL+/highMR+group was the shortest?p=0.008?,suggesting that MR interacts with its ligand-mannose in gastric cancer affect the progression and prognosis of gastric cancer patients.
