Purpose: 1.Protein microarray technology was used to screen out cytokines that may be related to lung cancer,and the relationship between differential factors and clinicopathological features of lung cancer and its diagnostic value were explored.2.Protein microarray was used to screen cytokines related to bone metastasis of lung cancer,and the correlation between related factors and bone metastasis of lung cancer was further analyzed by immunohistochemistry.Methods:1.Application of protein chips capable of detecting multiple cytokines at one time,screening differential factors in serum samples of 49 patients with lung cancer and20 healthy controls;statistically analyzing the relationship between differential factor expression and clinicopathological features of lung cancer and its diagnostic value.2.Further detection of serum samples from 9 patients with non-small cell lung cancer who only had bone metastases and 10 patients with non-small cell lung cancer without bone metastases by chip technique,looking for differentially expressed target factors.3.Immunohistochemical technique was used to further verify the expression of target factors in bone metastases of 12 patients with non-small cell lung cancer who only had bone metastases and 12 patients with non-small cell lung cancer without distant metastasis.Results: 1.The expression of various cytokines in lung cancer patients and healthycontrols was detected by protein chip.The expression levels of IGFBP-1,TNF RI,TNF RII,VEGF,PAI-1 and IL-6 in serum of lung cancer patients were significantly higher than those in lung cancer patients.The healthy control group(P<0.05);the expression level of BDNF in the serum of lung cancer patients was lower(P<0.05).2.The expression level of serum TNF RI was correlated with distant metastasis of lung cancer(P<0.05),but it was not correlated with pathological type,tumor size,lymph node metastasis and TNM stage of lung cancer patients(P>0.05).3.The area under the ROC curve for the diagnosis of newly diagnosed lung cancer patients with serum IGFBP-1,TNF RI,VEGF and TNF RII was 0.96,0.96,0.83 and0.80,respectively,and the sensitivity and specificity of the diagnosis were over 70%.4.In the serum of patients with non-small cell lung cancer,the expression of lipopoietin-2(LCN2)and platelet-derived growth factor-BB(PDGF-BB)was significantly higher in the bone metastasis group than in the boneless group.5.Immunohistochemical staining of 12 non-small cell lung cancer metastases with bone metastases only and 12 non-small cell lung cancers without distant metastasis showed that LCN2 and PDGF-BB were mainly distributed in the cytoplasm of tumor cells.And nuclear nuclei;LCN2 expression was positive in 10/12 bone metastases(83.33%),PDGF-BB expression was positive in 9/12 bone metastases(75.00%);LCN2 expression was positive in 1/12 primary lung cancer(8.33%),2/12 cases of PDGF-BB expression positive(75.00%).Statistical analysis showed that the expression of LCN2 and PDGF-BB in bone metastases was significantly higher than that in primary lung cancer(P<0.05).Conclusion: This study found that cytokines IGFBP-1,TNF RI,VEGF,TNF RII,PAI-1and IL-6 may be involved in the development and progression of lung cancer,including IGFBP-1,TNF RI,VEGF and TNF RII for early diagnosis of lung cancer.Provide a certain reference value;TNF RI is related to the distant metastasis of lung cancer;the expression of LCN2 and PDGF-BB is significantly higher in the serum of the bone metastasis group than in the non-bone metastasis group,and the tissue is also confirmed in the non-small cell lung cancer bone metastasis Significantly higher than the primary tumor,LCN2 and PDGF-BB may be predictors of bone metastasis in lung cancer.
