ObjectiveTo investigate the effects and mechanism of phloretin on the growth and apoptosis of human tongue cancer Tca8113 cells in vitro.MethodsThe effects of phloretin on the viability of Tca8113 were detected by MTT assay and clone formation assay.Heochst33342 staining and flow cytometry were performed to observe the effects of phloretin on the apoptosis of Tca8113 cells.Expression of the expression of Bax,Bcl-2,Cyt C,Caspase 3 protein in Tca8113 which were treated with phloretin can be measured by Western blotting.Real-time PCR was applied to detect the mRNA expressions of Bax,Bcl-2,Cyt C,Caspase 3.ResultsPhloretin inhibited viability and promoted apoptosis of human tongue Tca8113 cancer cells in a dose-dependent manner.Phloretin promoted the expression of Bax,Cyt C and Caspase 3 and inhibited the expression of Bcl-2.ConclusionsPhloretin can inhibit the proliferation of Tca8113 cells in vitro and promote its apoptosis,the mechanism may be related to the ratio of Bax/Bcl-2 expression,the enhancement of Cyt C expression and the activation of Caspase 3 expression.
