ObjectiveTo observe the effect of ursolic acid on the growth and migration of human tongue squamous cell carcinoma Tca8113 cells in vitro,and to explore its related mechanisms.MethodsThe effects of ursolic acid on the viability of Tca8113 were detected by MTT assay and colony formation assay.The effects of ursolic acid on the migration of Tca8113 cells at 0h,12 h and 24 h were detected by scratch test.Hoechst33342 staining and flow cytometry were performed to observe the effects of ursolic acid on the apoptosis of Tca8113 cells.Western blotting was used to detect the effects of ursolic acid on the expression of Bcl-2,Bax,Cleaved caspase-3,p-Fak and Fak.ResultsThe results of MTT assay showed that different concentrations of ursolic acid inhibited the activity of Tca8113 cells in different degrees,and the difference was statistically significant(P<0.05),and the cell growth inhibition rate was positively correlated with drug concentration.The results of colony formation showed that the cells in the medicated group grew slowly,containing few clones,and the cell survival was small at the end of the experiment,almost in the stationary phase,indicating that ursolic acid can inhibit the activity of Tca8113 cells;the results of the scratch test showed The scratches in the experimental group were significantly wider than the control group,and the mobility was positively correlated with the action time,indicating that ursolic acid could inhibit the migration of Tca8113 cells;The results of Hoechst33342 and flow cytometry showed that ursolic acid was Apoptosis increased gradually after 0h,12 h,and 24 h,indicating that ursolic acid can induce apoptosis of Tca8113 cells(P <0.05)in a time-dependent manner.The mechanism lies in that ursolic acid increased apoptosis of Tca8113 cells by inhibiting the expression of Bcl-2 and increasing the expression of of Bax,resulting in the increasing expression of Cleaved caspase-3;And inhibited migration of Tca8113 cells by decreasing the expression of p-Fak and Fak.ConclusionUrsolic acid can significantly inhibit the growth and migration of tongue cancer cell line Tca8113 and promote its apoptosis.The mechanism may be related to the regulation of Bcl-2/Bax apoptosis pathway and inhibition of Fak activation.
