Objective: To observe the healing effect and explore the mechanism of Haifu powders on the proliferation of new blood vessels on wound surface.Methods: Ninety SD rats were operated to establish A model of open back wound.After successful modeling,they were randomly divided into grouping A(model group),group B(control group)and group C(experimental group)with 30 rats in each group.The wounds of rats in different treatment groups were changed daily: rats in group A were rinsed with normal saline,rats in group B were applied with vaseline gauze,and rats in group C were applied with Haifu powder.The activity of rats on the first,third,seventh and fourteenth days after surgery was observed,the wound healing area was recorded,and the wound healing rate was calculated.The area ratio of neovascularization was observed and calculated by staining and sampling of wound tissue in different phases.The content of VEGF in wound tissue was determined by immunohistochemistry to prove that Haifu powder had positive effect on wound healing.Results: 1.There was no significant difference in wound healing rate of groups A,B and C on the first and third days after surgery(P > 0.05).On day 7 and day 14,the wound healing rate of group B and C was higher than that of group A,with statistically significant difference(P < 0.05).2.HE stained observation showed that the proliferation of inflammatory cells in the wound tissue of rats in group A at different time points after dressing change was more obvious than that in group B and C,which gradually decreased with time.The wound fibroblasts and granulation tissue of rats in groups A,B and C increased gradually,and those in groups B and C increased significantly compared with those in group A.New capillaries increased first and then decreased.The vascular area ratio of group C was better than that of group A and group B(P < 0.01),and that of group B was better than that of group A(P < 0.05).3.Immunohistochemical results showed that the expression of VEGF in the wound tissue of rats in group C was higher than that in groups A and B,with A significant difference(P < 0.05).The expression of VEGF first increased and then decreased to the passage of healing time,and reached A peak on day 7.Conclusion: 1.Haifu powder can promote the wound healing in the back of rats,thus improving the healing rate and quality.2.The effect of Haifu Powder on wound in rats can regulate the expression of VEGF in the wound tissue,thereby affecting the proliferation and differentiation of capillary vessels on the wound and accelerating the healing.
