Forsythiae Fructus is the dried fruit of the Forsythia Suspensa(Thunb.)Vahl(Oleaceae).Although the current national standards,ministry edition standards and the processing standards of Forsythiae Fructus in various provinces and municipalities as well as the enterprise standards have stipulations on the processing technology of Forsythiae Fructus,there is no unified standard for the processing technology of prepared slices of Forsythiae Fructus.The processing technology of prepared slices of Forsythiae Fructus can affect the quality of the slices and directly affect the medication safety and therapeutic effectiveness of patients.Therefore,this experiment studied the processing technology of prepared slices of Forsythiae Fructus,determined that carpopodium should be removed in processing of prepared slices of Forsythiae Fructus,what can better standardize the processing technology of prepared slices of Forsythiae Fructus.Up to now,there is no recognized national or industrial standard for the grade of prepared slices of Forsythiae Fructus.The grade standard for prepared slices of Forsythiae Fructus by combining traditional grading method with modern scientific and technological methods.Through the correlation between appearance traits and internal quality of prepared slices of Forsythiae Fructus was analyzed,and the grade-related appearance traits were determined.By using modern statistical methods,the correlation between grade standard and effective ingredients of prepared slices of Forsythiae Fructus was analyzed,and the components positively correlated with grade standard were selected.The quality grade of prepared slices of Forsythiae Fructus was evaluated comprehensively by combining grade standard with chemical quantitative index.The results reveal the intrinsic relationship between prepared slices of Forsythiae Fructus and quantitative indicators of chemical components,and provide experimental basis for the formulation of grade standards of prepared slices of Forsythiae Fructus.The main contents of this paper are as follows:1.The processing technology of processed Forsythiae Fructus was studied.It was found that there was little difference in water content between the two groups.The content of alcohol extract,forsythiaside and forsythiaside A were higher than those with handle Forsythiae Fructus.Comprehensive evaluation showed that the handle should be removed in the processing of prepared slices of Forsythiae Fructus.2.The quality of 32 batches of Green Forsythiae Fructus decoction pieces and 17 batches of Grown Forsythiae Fructus decoction pieces were analyzed.The quality of Forsythiae Fructus decoction pieces was identified from the aspects of properties,microscopy and thin layer.The results showed that all the samples collected were genuine Forsythia suspensa.According to the method of Chinese Pharmacopoeia of 2015 edition,impurities,moisture,total ash,alcohol-soluble extract and water-soluble extract of Forsythiae Fructus were determined.The impurity contents of 32 batches of Green Forsythiae Fructus and 17 batches of Grown Forsythiae Fructus all meet the Pharmacopoeia standards.Water content ranged from 5.8% to 9.5% in 32 batches of Green Forsythiae Fructus and from 3.3% to 10.6% in 17 batches of Grown Forsythiae Fructus.The total ash content of Green Forsythiae Fructus and Grown Forsythiae Fructus is less than 4%,which increases the determination of acid-insoluble ash.It is stipulated that the total ash content and acid-insoluble ash content of different grades of Forsythiae Fructus should be less than 4.0% and 1.0% respectively.The content of alcohol extract in 32 batches of Green Forsythiae Fructus decoction pieces is 26.9%-48.3%.The content of alcohol extract in 17 batches of Grown Forsythiae Fructus decoction pieces is 7.5%-19.1%.It is stipulated that the content of alcohol extract in different grades of Green Forsythiae Fructus decoction pieces is not less than 30%,and that in Grown Forsythiae Fructus decoction pieces is not less than 16%.The determination of water-soluble extract was increased.According to the determination results,the water-soluble extract content of different grades of Green Forsythiae Fructus decoction pieces was more than 15%,and that of Grown Forsythiae Fructus decoction pieces was more than 8%.The contents of forsythiaside A and lignans in 32 batches of Green Forsythiae Fructus decoction pieces and 17 batches of Grown Forsythiae Fructus decoction pieces were determined.The contents of forsythiaside A in Green Forsythiae Fructus decoction pieces ranged from 0.25% to 9.67%,forsythiaside A in Grown Forsythiae Fructus decoction pieces ranged from 0.15% to 4.71%,and the contents of pineolin-beta-D-pyranoside in Green Forsythiae Fructus decoction pieces ranged from 0.26% to 0.87%,and the contents of pineolin-beta-D-pyranoside in Grown Forsythiae Fructus decoction pieces ranged from 0.07% to 0.70%.The range of determination of forsythiaside in Green Forsythiae Fructus decoction pieces is 0.11%-0.89%.The range of determination of forsythiaside in Grown Forsythiae Fructus decoction pieces is 0.04%-0.28%.The range of determination of forsythiasin in Green Forsythiae Fructus decoction pieces is 0.03%-0.55%.The range of determination of forsythiasin in Grown Forsythiae Fructus decoction pieces is 0.04%-0.17%.Qualified slices of Forsythiae Fructus will be classified into different grades,which will provide data support for the study of grade standards of F Forsythiae Fructus slices.3.The relationship between the appearance and internal quality of the slices was studied,and the size of the slices was digitalized.The correlation between the size of the slices and the effective components was analyzed by SPSS software.The results showed that there was no correlation between the size of the prepared slices of Forsythiae Fructus and the content of the effective components.Therefore,when dividing the grade of the slices of Forsythiae Fructus,the size of the slices of Forsythiae Fructus was not taken as the index of dividing the grade.The correlation between the color of Forsythiae Fructus pieces and the content of the main active ingredients was studied.The results showed that the correlation coefficient between the content of terpinin-beta-D-pyran glucoside and a was-0.554(P < 0.01),and the correlation coefficient between forsythia with L and a were-0.421(P < 0.01)and-0.563(P < 0.01),while the correlation coefficient of forsythiaside A and a was-0.753(P < 0.01).Using modern statistical methods,the correlation between the grade standard of Forsythiae Fructus pieces and the quantitative indexes of four chemical constituents was analyzed.The results showed that there were significant positive correlations between the three indexes of pine resin-beta-D-pyran glucoside,forsythiaside,forsythiaside A and clustering categories.Therefore,the three indexes were selected as the quality correlation index components of Forsythiae Fructus pieces,and finally combined with traditional methods.The grade standard of Forsythiae Fructus pieces was established by the appearance index of grade method and the physical and chemical index of modern grading method.4.ELISA was used to detect the anti-inflammatory effect of different grades of Forsythia Fructus extracts in vitro.The results showed that the anti-inflammatory effect of the first grade was greater than that of the second grade,and the anti-inflammatory effect of the second grade was greater than that of the third grade.Therefore,the rationality of grade division is verified.
