1 Purpose Through the determination of Qi Huang Decoction(QH)by High Performance Liquid Chromatography(HPLC),the pharmacodynamic material basis of QH on SCF/c-kit signaling pathway was revealed.Then the Wistar rat model of gastrectomy was established and QH was infused into the small intestine to observe the effect of QH on the recovery of intestinal function after gastrectomy,and the role of SCF/c-kit signaling pathway in the recovery of intestinal function after gastrectomy.The mechanism of QH on the recovery of intestinal function after gastrectomy was preliminarily explored.2 methods2.1 The fingerprints of different batches of Qihuang Decoction were constructed by high performance liquid chromatography(HPLC),and the corresponding common peak data were obtained.Similarity evaluation software(2004A)was used for processing and analysis.2.2 A total of 60 rats were randomly divided into control group,standard enteral nutrition group(EN),standard enteral nutrition+Qihuang Decoction group(EN+QH)and standard enteral nutrition + Qihuang Decoction group + c-kit inhibitor group(AG-1296)(EN+QH+AG-1296).The blank control group was given no treatment,free drinking water,no enteral nutrition and traditional Chinese medicine.In the sham-operation group,only mid-abdominal incision and suture were performed,but no gastrectomy was performed.No enteral nutrition and traditional Chinese medicine were given after operation.All the other three groups underwent gastrectomy and received standard enteral nutrition.The EN+QH group was given 10 g/(kg.d)Qihuang decoction,and the EN+QH+AG-1296 group was given 10 mg/kg/d Ag-1296 on the basis of EN+QH group.After treatment,the electrophysiology and propulsive rate of small intestine were measured,the content of serum SCF was determined by ELISA,and the muscle layers of small intestine were isolated.The number and distribution of SCF and c-kit double positive cells(ICC)were analyzed by indirect immunofluorescence,and the expression levels of SCF and c-kit m RNA and protein in different groups were detected by fluorescence quantitative PCR and Western blot.3 Results3.2 It is shown that Qihuang decoction can significantly improve the rhythm of small intestine peristalsis in gastric resected rats,while c-kit inhibitor AG-1296 can inhibit the function of Qihuang decoction.Compared with EN group(30.5±4.0%),EN+QH group(39.36±2.9%)significantly increased intestinal propulsion rate(P=0.034);EN+QH+AG-1296 group(24.3±1.7%)and EN+QH Compared with the EN group,the serum SCF concentration in the EN+QH group was significantly higher than that in the EN group(P=0.014);the EN+QH+AG-1296 group was compared with the EN+QH group in the serum SCF.The concentration of SCF decreased significantly in the EN+QH group compared with EN(P=0.017)and EN+QH+AG-1296(P=0.005).The C-kit IOD value in the EN+QH group was significant.Higher than EN group(P=0.012)and EN+QH+AG-1296 group(P=0.0054);the expression level of SCF m RNA in EN+QH group was significantly higher than that in EN group(P=0.034)and EN+QH+AG-1296 group(P=0.008);the expression level of c-kit m RNA in EN+QH group was significantly higher than that in EN group(P=0.028)and EN+QH+AG-1296 group(P=0.006);EN+QH group SCF protein The expression level of the c-kit protein in the EN+QH group was significantly higher than that in the EN group(P=0.021)and EN+QH+AG-1296(P=0.004).EN+QH+AG-1296 group(P=0.005).4 Conclusion QH quality can be controlled.QH can promote the recovery of small intestinal motility in rats after gastrectomy.The mechanism of QH promoting the recovery of small intestinal function in rats after gastrectomy may be through up-regulating the expression of SCF/c-kit signaling pathway and increasing the number of ICC.
