Objective: We builded gastrectomy trauma model by animal experiments, in theintervention of infusion Qi Huang Decoction in the small intestine, we observed theinfluence of Qi Huang Decoction on intestinal lymphocytes homing process of gutmucosa after gastrectomy. Layed the foundation for solving modern critical illness oftraditional Chinese medicine.Methods: These rats were randomly divided into three groups:(1) sham group: Onlysuturing after abdominal midline incision, without gastrectomy. Free water to eat afterthe operation, without giving enteral nutrition and Chinese traditional medicine.(2)control group: After establishment the gastrectomy trauma model, placed them inmetabolic cages, they can free activities in the cages after waking, fasted on the day ofsurgery. Starting at the first day after operation, giving them enteral nutrition bymicrosyringe pump. We choosed nutrison as enteral nutrition. Dripped of saline6mlbefore dripping nutrient solution.(3) Qi Huang Decoction group: intravenous injectionof nutritional formulations, heat, methods and treatments was the same as the simplyenteral nutrition group, meanwhile dripping Qi Huang Decoction. Before infusion of thenutrient solution can be given vegan Qi Huang Decoction6ml, injected into the smallintestine uniformly and continuously for a week.observation of samples and testing methods after treatments:(1) Detection the number of lymphocyte homing: After markered the sodiumchromate that comes from lymph fluid obtained by mesenteric lymph drainage, anddetective the stability of radioactivity counts with γ counter. Infused slowly the51Cr thathas a stability radiation counting into the body from the femoral vein. After one hour,detected and calculated the total number of lymphocytes that51Cr labeled in theintestinal mucosa and lymph tissue specimens and with γ counter.(2) Observation of T lymphocytes in lymph: Detected αβTCR-CD3+T cells and thesubsets of CD4+、CD8+T cells through directly drainaged the lymph fluid in intestinallymphoid tissues by cytometry method. (3) Isolated and detection PP knot IgA+B cells: PP knot was separated, paraffinsections, detected IgA+B cells by immunohistochemistry.Data Collection: The excitation light of flow cytometry was488nm hydrogen ionlaser, receiver channel FS (forward scattering), SS (side scatter), observed subsetschanges of CD3+T cells, CD4+T, CD8+T cell, analyzed the data using theCell-quest software.Statistical methods: Application SPSS17.0software for statisticalanalysis.Between groups of sample data to the f test, by using the Levene, if variance,with single factor analysis of variance design, further than using t test sample;If thevariance is not neat, namely using nonparametric sample test.With the mean±standarddeviation (x±s) show the data from experiment, To determine differences havestatistical significance by P>0.05.Results:1The number of lymphocytes homing(The percentage of51cr accounted fortotal volume in the three kinds of organization including rats Peyer’s patches,mesenteric lymph nodes, small intestinal diffuse lymphoid tissue):(1)Peyer’s patches:Control group was4.93±0.50, the control group was2.85±0.37, percentage in thecontrol group were significantly lower than the control group, has obvious significantdifference (P﹤0.01).Qi Huang Decoction group was3.24±0.28, the percentage lowerthan the control group, has obvious significant difference (P﹤0.01);Qi HuangDecoction group and control group, compared with Qi Huang Decoction grouplymphocyte homing percentage is better than that of control group, with significantdifference (P﹤0.05).(2) mesenteric lymph nodes:control group was3.85±0.28, thecontrol group was2.70±0.54, the control group compared with control group, thepercentage is lower than the control group, has obvious significant difference (P﹤0.01).Qi Huang Decoction group was3.53±0.40, compared with control group, nosignificant difference (P>0.05);Qi Huang Decoction group compared with control group,the percentage is higher than the control group, has obvious significant difference (P﹤ 0.01).(3) the intestinal diffuse lymphoid tissue: there were no significant differencescompare between the three groups, with no statistical significance (P>0.05).2Lymphocytes homing T cell number(CD3+T cells and their subsets CD4+,CD8+T cells changes in rats lymph: After modeling rats a week): after1week, thedrainage of lymph cells by the Flow Cytometer,(1)CD3+T cells: the control groupwas77.24±3.86, the control group was67.59±3.99, the control group were lower thancontrol group, has obvious significant difference (P﹤0.01).Yellow stilbene decoctiongroup was74.04±3.40, compared with control group, with significant difference (P <0.05);Qi Huang Decoction group compared with control group, has obvious significantdifference (P﹤0.01).(2) the CD4+T cells: the control group was54.42±4.09, the controlgroup was50.31±3.22, the control group were lower than control group, has obvioussignificant difference (P﹤0.01).Qi Huang Decoction group was51.11±2.98, comparedwith control group, with significant difference (P<0.05);Qi Huang Decoction groupcompared with control group, has no significant difference, no statistical significance (P>0.05).(3) CD8+T cells: control25.04±3.94,20.44±5.13in the control group, controlgroup were lower than control group, with significant difference (P﹤0.05).Qi HuangDecoction group was21.95±6.01, compared with control group and the control group,no obvious difference, has no statistical significance (P>0.05).3Lymphocytes homing B cells (The number change of IgA+B cells in PPjunction of the small intestine of rats):Control group was69.74±17.682, the controlgroup was41.29±16.623, the control group compared with control group, withsignificant difference (P<0.01);Qi Huang Decoction group was57.08±10.908,compared to control group, compare the difference was statistically significant (P <0.05);Qi Huang Decoction group was higher than control group, the more significantdifferences (P <0.01).Conclusion: Traditional Chinese medicine (TCM)QI HUANG decoction bypromoting gastric bowel lymphocytes homing after surgery, so as to protect theintestinal mucosal immune barrier, the prevention and treatment of bacterial translocation, helps reduce the surgical trauma after enterogenic infection, SIRS andMODS.
