Objective: These studies intend to detect the changes of PGC-1?/ERR-?/VEGF expression after establishing the skin defect model and the influence that VSD treat on them,and to explore the angiogenesis mechanism of VSD promotes wound repair.Methods:1.Randomly selected 6 experimental New Zealand rabbits,established acute full-thickness skin defect wound at the back of the rabbit on both sides of the spine.2.Each New Zealand rabbit was randomly selected as the VSD treatment group and the control group on both sides of the back.3.Tissue changes at different time points during wound healing were observed by naked eye and HE staining.Skin wound tissues at 6h,12 h,1st,2nd,3rd,7th and 11 th day after surgery were collected,and the temporal and spatial expression of PGC-1? and ERR-? mRNA were detected by qRT-PCR.Western Blot was performed on some wound tissues 1st,2nd,3rd,7th and 11 th day after surgery to detect the protein expression levels of PGC-1? and VEGF.The expression of ERR-? protein was detected by ELISA.Results:1.Postoperative wound gross observation: compared with the control group,wound surface in the VSD group was dryer,moister,and brighter red,with rarely exuded necrotic materials.Fresh granulation tissue was visible on the second day after surgery in the VSD group and it was more obvious on the 7th and 11 th days.2.Histological manifestations: HE staining was performed on the 1st,2nd,3rd,7th and 11 th day after operation.It was found that inflammatory cell infiltration of different degrees was observed in both groups on the 1st day after surgery.On the second day after surgery,newborn capillaries were observed in the VSD group,while more necrotic substances were found in the control group.On the 3rd day,capillaries in VSD group were significantly more than those in control group.On the 7th day,granulation tissue became thicker and a large number of newborn capillaries could be seen in VSD group.In VSD group,the granulation tissue matured and a small amount of capillaries were observed at 11 days after operation.In control group,a large number of new capillaries were observed.3.The results of qRT-PCR showed that the mRNA expression levels of PGC-1? and ERR-? in VSD group were significantly higher than those in control group at 6h,12 h,1st,2nd,3rd,7th day after operation,and the difference was statistically significant(p<0.05).On the 11 th day,the expression levels of PGC-1? and ERR-? in VSD group were significantly lower than those in control group(p<0.05).The mRNA expression of PGC-1? and ERR-? in VSD group increased slightly with time,and showed a high level on the 3rd and 7th day,then began to decline gradually.The expression levels of PGC-1? and ERR-? in control group increased gradually with time.4.Western Blot results showed that the expression of PGC-1? protein in VSD group was higher than that in control group on the 1st,2nd and 3rd day after operation,with statistical significance(p<0.05).On the 7th and 11 th day,the expression of PGC-1? protein in VSD group was higher than that in control group,with no statistical significance(p>0.05).The expression level of VEGF protein in VSD group was higher than that in control group at 1,2,3 and 7 days after operation(p<0.05).There was no significant difference between the two groups at 11 th days.5.ELISA results showed that the expression of ERR-? in VSD group was higher than that in control group on the 1st,2nd and 3rd day after operation,and the difference was statistically significant(p<0.05).There was no significant difference in the expression of ERR-? on the 7th and 11 th day between the two groups.Conclusion:1.PGC-1?/ERR-? is involved in the natural healing process of wound repair after injury,which is time-dependent.2.Up-regulating the expression of PGC-1?/ERR-?/VEGF may be the mechanism that VSD promotes wound angiogenesis after injury.
